Antioxidant Potential of Dictyophora indusiata

Dictyophora indusiata was purchased in Nanping Shengli Market. After the Dictyophora indusiata is air-dried, it is fully ground into powder by a high-speed pulverizer. DPPH (1,1-diphenyl-2-picroyl), ABTS (total antioxidant activity kit), PMS, NATB were purchased from Sigma Company in the United States (America). EDTA, trichloroacetic acid, ascorbic acid, Tris-hydrochloric acid buffer, H2O2 absolute ethanol, chloroform, sodium chloride, concentrated sulfuric acid, chloroform, n-butanol were purchased from Shanghai Chemical Reagent Factory (Shanghai, China). Methanol, trifluoroacetic acid, sodium hydroxide and sodium acetate trihydrate were purchased from sigma (Sigma–Aldrich, St. Louis, MO, USA). The chemicals and solvents used in this study were of analytical grade.

Plasma miRNA Extraction from EDTA-Preserved Blood

Blood was drawn from the above-mentioned subjects recruited into this study. About 1 ml of peripheral whole blood were collected from each individual into free-enzyme-containing ethylenediaminetetraacetic acid (EDTA) tubes. Following the protocol provided by the producers, the blood was collected into EDTA tubes for centrifugation at 12,000 g for 10 min at 4 °C. Finally, the precipitate was collected into 1.5 ml Eppendorf (EP) tubes and stored at −80 °C for further analysis.
After the preparation of the plasma, the miRNAs were extracted using the TRIZOL method, following the producers’ protocol. We add 0.75 m L of TRIZOL reagent and 0.2 m L of chloroform (Shanghai Chemical Reagent Co., Ltd, China) to the samples and mix well, let it stand for 3–5 min; centrifuge for 10 min at 4 °C and 12,000 g centrifugal force; aspirate the supernatant and transfer it to another EP tube, then add 0.5 m L of isopropanol and mix well. Incubate at −20 °C overnight; centrifuge again at 4 °C and 10,000 g for 10 min, discard the supernatant, then add 1 m L of 75% ethanol, mix well, and continue centrifugation at 4 °C and 10,000 g for 10 min; then air dry at room temperature, add 30 μL of DEPC-treated dd H2O water to dissolve the RNA, and store at −80 °C. Storage.

Ginkgo Fatty Acid Profiling

All fatty acids (C5–C30) standards were of analytical grade and purchased from Sigma-Aldrich (St. Louis, MO, USA). HPLC grade acetonitrile (ACN) was purchased from Yucheng (Shandong Province, China). N,N-Dimethylformamide (DMF), potassium carbonate (K2CO3), pyridine, and chloroform were of analytical grade obtained from Shanghai Chemical Reagent Co. (Shanghai, China). Toluene-4-sulfonic acid 2-(2-thiophen-2-yl-phenanthro[9,10-d]imidazol-1-yl)-ethyl ester (TSTPE) was synthesized in our lab as described in the synthesis section and Fig. 1. Water was purified on a Milli-Q system (Millipore, Bedford, MA). All other reagents used in this study were also of analytical grade unless otherwise stated. Ginkgo nut and ginkgo leaf samples were collected from Shandong province, Jiangsu province and Henan province of China.

Synthesis and Analysis of AEAO

Processed tea samples were bought from Hangzhou West Lake Longjing Industry Co.(June, 2011), Ltd. and Guizhou Fanjin Industry Co., Ltd.(July, 2011).
