Glycemic Markers in Murine Metabolism

Check if the same lab product or an alternative is used in the 5 most similar protocols
Glycated hemoglobin (HbA1c) has been validated as a reliable marker to measure glycemic control in mice (Feng et al., 2019 (link); Han et al., 2008 (link); Dan et al., 1997 (link)). Whole blood HbA1c and plasma levels of TAG and prolactin were measured. HbA1c was measured in whole undiluted blood collected in EDTA tubes using a commercial HbA1c assay kit (Crystal Chem, Elk Grove Village, IL, USA). Plasma TAG was measured by preparing a 1:5 dilution using the Cayman Chemical kit. Plasma prolactin was measured in a 1:20 dilution of plasma using a commercially available ELISA kit (ab100736, Abcam, MA, USA). All assays were performed following the manufacturers’ instructions.

Plasma Prolactin Quantification in Shank2 Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected via cardiac puncture from 8‐ to 10‐week‐old Shank2+/+ and Shank2−/− dams (postnatal day 0) in tubes containing EDTA (2 mg/ml) as an anticoagulant and aprotinin (0.6 trypsin inhibitory units TIU/ml of blood) was added in order to inhibit the activity of proteinases. Blood was immediately centrifuged at 1,600 × g for 15 min at 4°C. The resulting supernatant (plasma) was collected. Prolactin concentration of the plasma was determined using an enzyme‐linked immunoassay kit (Abcam, ab100736), according to the manufacturer’s instructions.

Acute Sleep Deprivation Serum Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immediately after acute sleep deprivation, mice were anaesthetized with 2–5% isoflurane and whole blood was collected by inferior vena cava puncture and coagulated at room temperature for 1 h. The serum was isolated by centrifugation at 6000 rcf, 10 min, at 4°C, and stored at −80°C until use. Serum corticosterone was quantified by a mouse corticosterone ELISA kit according to manufacturer instructions (ADI-900-097; Enzo Life Science, Farmingdale, NY, USA). Serum prolactin level was quantified by a mouse prolactin ELISA kit according to the manufacturer’s instructions (ab100736; Abcam, Waltham, MA, USA).

Serum Prolactin Quantification After U-69593 or Nalfurafine

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected 30 minutes after a single U-69593 or nalfurafine administration for evaluation of serum PRL levels in different cohorts. Mice were deeply anesthetized with isoflurane (2%) and whole blood was collected by inferior vena cava puncture and coagulated at room temperature for 1 hour before isolating serum by centrifugation at 6,000 rcf, 10 minutes, 4°C. Serum samples were collected and stored at −80 °C until use. Serum prolactin was quantified by a mouse prolactin ELISA kit according to manufacturer instructions (ab100736; Abcam, Cambridge, UK).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
