Analytical Validation of Novel Psychoactive Substances

Reference materials were used for the validation and stability analysis. All chemicals and solvents used were of analytical grade. Methanol was purchased from Fisher Chemicals (Loures, Portugal), formic acid was purchased from Merck (Darmstadt, Germany), and ethanol was purchased from Merck (Darmstadt, Germany). 4-methylmethcathinone (mephedrone), butylone, pentylone, eutylone, ephylone, 4-methyl-α-ethylaminopentiophenone (4-MEAPP), 4-methoxyamphetamine (PMA), para-methoxymethamphetamine (PMMA), deschloroketamine (DCK), mephedrone-D3, butylone-D3, 3,4-methylenedioxyamphetamine-D5 (MDA-D5), and ketamine-D4 were purchased from Cerilliant (Texas, USA). 2-fluorodechloroketamine (2-FDCK) was purchased from Cayman Chemicals (Ann Arbor, MI, USA).

Quantitative Analysis of Ketamine and Metabolites

ketamine dosing solution was purchased from Putney, Inc. (Portland, MA) and either used as directed or aseptically diluted using sterile saline to a final concentration of 25 mg/mL. The concentration of the ketamine dosing formulations was verified using LC-MS as described below. Atropine was purchased from Sigma Aldrich (St. Louis, MO) and prepared at a concentration of 15 mg/mL in sterile saline. The pH of the vehicle was acidified with dilute hydrochloric acid to increase solubility before being neutralized to pH 7.2 with dilute sodium hydroxide. The 2-PAM used was purchased from Sigma Aldrich (St. Louis, MO) and prepared at 50 mg/mL in sterile saline and sterile filtered. Midazolam (5 mg/mL) was purchased from Hospira (Lake Forest, IL) and used without modification.
The analytical reference standards for ketamine, norketamine, ketamine-d4, and norketamine-d4 in methanol were obtained from Cerilliant (Round Rock, TX). Multiple lots of K3EDTA rat plasma and brains were obtained from Bioreclamation (Westbury, NY). Purified water was collected from an in-house Labconco (Kansas City, MO) Water Pro Plus system. Glacial acetic acid was obtained from JT Baker (Center Valley, PA). Ammonium hydroxide was obtained from Sigma Aldrich (St. Louis, MO). LC-MS-grade water and acetonitrile were obtained from EMD (Billerica, MA). Formic acid was obtained from Thermo Scientific (Waltham, MA).

Extraction and Quantification of Bioactive Compounds

Acetone, ligroin, sodium chloride (NaCl), sodium dodecyl sulfonate (SDS), octly β-D-glucopyranside (OG), myoglobin and lysozyme were purchased from Sigma (St. Louis, MO). Gramincidin D (from bacillus breris) was purchased from International Lab (USA).
Ketamine was purchased from Alfasan (Woerden, Holland). Methanol, acetonitrile and chloroform were purchased from Tedia (Fairfield, OH). Ketamine-D4 was purchased from Cerilliant (Round Rock, Texas). Petroleum was purchased from Fisher Scientific (Pittsburgh, Pa.). Formic acid was purchased from Fluka (Buchs, Germany). Ethyl acetate was purchased from Acros (Fairlawn, NJ). Ginsenoside Rc was purchased from Shanghai Tauto Biotech (Shanghai, China). The urine sample was collected from a healthy male volunteer. Herbal medicines Fruit of Schisandra sphenanthera (FSS) and Fruit of Schisandra chinensis (FSC) were purchased from pharmacy stores in Hong Kong. Fresh spinach leaves were purchased from a supermarket in Hong Kong. Extracts of spinach leaves, FSS and FSC, were prepared as described previously [38] . TLC plates (Model: Alugram Sil G/UV254) were purchased from Macherey-Nagel (Düren, Germany).
