Genetics of Drosophila Learning Behavior

The following strains were used: y,w and CS were used as the control; y, w, dlgXI-2; y, w, dlgS975; y, w, dlgA40.2; (Bloomington Stock Center, Bloomington, IN, USA). All of the strains were backcrossed six-fold with Canton-S to standardize the genetic background (Cantonization). For the adult associative olfactory learning experiments, the strains used were: W(CS); Ok107-GAL4 (kind gift from Dr. Jorge Campusano); UAS-RNAi-dlg (Vienna Drosophila Resource Center, AT); UAS-dsRNA-dlg1; Orco-GAL4; GH146-GAL4; Tenm-GAL4;117y-GAL4; C305A-GAL4; Elav-GAL4 (Bloomington Stock Center, Bloomington, IN); Ok107-GAL4; UAS-DlgS97 (Kind gift from Dr. U. Thomas). For the dNR1 and dNR2 overexpression, UAS-GFP-CD8-dNR1 and UAS-GFP-CD8-dNR2 flies were used. All of the flies were grown at 22 °C.

Drosophila Rearing and Genetics Procedures

Drosophila melanogaster flies were raised on standard medium (water, cornmeal, soy bean, agar, molasses, yeast, preservative methyl-4-hydroxybenzoate) at 25 °C, 60% humidity and 14 h/10 h light–dark cycles. Canton-S (CS) served as wild-type Drosophila strain in this study. The following colleagues kindly provided additional stocks: D. Armstrong (189Y-GAL4), M. Heisenberg (wild-type Canton-S, rut1, and dnc1), T.J. Hirsh (Trh493-GAL4), Y. Rao (5HT-1A-GAL4), H. Scholz (UAS-TNT), P. Shen (UAS-npf), H. The following lines were obtained from the Bloomington Stock Center: Or83b-GAL4 (#2681), c305a-GAL4 (#30829), c739-GAL4(#7362), npf-GAL4 (#25681), 5HT-1B-GAL4 (#27637), UAS-NaChBac (#9469), UAS-5HT-1ARNAi (#25834 and #33885), UAS-5HT-1BRNAi (#27634 and #54006), UAS-CD4::tdTomato (#35837) and UAS-mCD8::GFP (#5130).

Drosophila Husbandry and Genetic Tools

Flies (Drosophila melanogaster) were generally kept at 25°C and subjected to a 12 h light – 12 h dark cycle. A cornmeal medium supplemented with yeast, fructose and molasses was used to rear the flies. Canton-S was used as wild-type (courtesy of R. Genetic Manipulation of Plum in Drosophila
