Cholesterol Metabolism Regulation Assay

Dulbecco’s modified eagle medium (DMEM-F12), fetal bovine serum (FBS), penicillin and streptomycin solution, and trypsin-ethylenediaminetetra acetic acid were purchased from Life Technologies (Eugene, OR, USA). Phosphate buffered saline (PBS) was obtained from Biochrom AG (Berlin, Germany). HMG-CoA reductase assay kit was purchased from Sigma Chemical Co. (St. Louis, MO, USA). (1α,2α(n)-3H) Cholesterol (specific activity, 49 Ci/mmol) was purchased from Perkin-Elmer (Wellesley, MA, USA). NPC1L1 antibody was bought from Novus Biologicals (Littleton, CO, USA). Ezetimibe was purchased from Schering-Plough Research Institute (Kenilworth, NJ, USA). Folin-Ciocalteu reagent, sodium bicarbonate, 1,2-di-O-lauryl-rac-glycero-3-glutaric acid 6’-methylresorufin ester, taurocholic acid sodium salt hydrate, glycodeoxycholic acid, taurodeoxycholic acid, and hydrazine hydrate solution were received from Sigma-Aldrich Co. (St. Louis, MO, USA). All other chemical reagents used in this study were obtained from commercial sources.

Bile Extract and Conjugated Salts Protocol

Bile stock solution (12.5%) was prepared by dissolving crude porcine bile extract (Sigma, B8631) in RPMI or YPD. Insoluble components were removed by centrifugation at 12000 ×g, the supernatant filter sterilized and stored at 4°C in the dark. Sodium taurodeoxycholate hydrate (Sigma, T0875), taurocholic acid sodium salt hydrate (Sigma, T4409), sodium glycocholate hydrate (Sigma, G7132), and unconjugated bile salts (UBS) (Sigma 48305, 50% cholic acid sodium salt and 50% deoxycholic acid sodium salt) were dissolved at 100 mg/ml in either YPD or RPMI, filtered and used as stock solutions.

Rapid DNA Extraction from Bacteria and Protozoa

A DNeasy Blood and Tissue Kit (Qiagen, Hilden, Germany) was used as the spin column reference method. DNA of S. aureus and E. coli was extracted following the manufacturer’s instructions for Gram-positive and -negative bacteria, respectively. The only modification for S. aureus was the inclusion of bead beating as the pellet was resuspended in 400 µL PBS and transferred into a Soil Grinding SK38 Precellys Lysing Tube (Bertin, Montigny-le-Bretonneux, France). The suspension was mixed in Precellys 24 Tissue Homogenizer (Bertin, Montigny-le-Bretonneux, France) at 6500 rpm for one minute. A total of 200 µL of the cell suspension was treated as described in the manufacturer’s basic protocol. For C. parvum, 100 µL of 1.2% taurocholic acid sodium salt hydrate (Sigma Aldrich, St. Louis, MO, USA) was added to the cell suspension followed by incubation at 37 °C for 120 min. The suspension was pelleted (13,000× g, 5 min) and resuspended in 80 µL PBS, 100 µL ATL Buffer (Qiagen, Hilden, Germany) and 20 µL PK. The procedure was continued as the recommendation by the manufacturer’s protocol for Gram-negative bacteria. This was considered a “spin column” reference method.

Bile Acids and Macrophage Inflammatory Response

Plasma cytokine levels of IFN-γ, IL-2, IL-6, IL-4, IL-1β, TNFα, and IL-10 were examined using a mouse multi-spot proinflammatory panel kit from Meso Scale Discovery (MSD), according to the manufacturer’s protocol.
For determining the effect of bile acids on IL-6 expression and cell viability in RAW 264.7 murine macrophage cells, after 24 h from splitting 3000 cells into one well of 96-well plates, cells were pretreated with taurocholic acid sodium salt hydrate (Sigma), sodium tauroursodeoxycholate (Selleck), and taurochenodeoxycholic acid (Selleck) (5, 25, and 125 μM) for 2 h. Subsequently, the cells were stimulated with 50 ng/mL lipopolysaccharide (LPS) for 12 h. Following this, the concentration of IL-6 in the supernatant was measured by IL-6 mouse ELISA kit (Thermo Fisher) and cell viability was measured by TACS XTT cell proliferation assay kit (Trevigen, Inc.).

Adzuki Beans Lipid Metabolism Study

(Beijing, China). Other chemicals, such as sodium dihydrogen phosphate dehydrate, disodium hydrogen phosphate dodecahydrate, were of analytical grade. Macroporous adsorptive resins AB-8 was supplied by Nankai University, and polyamide resin was purchased from Sinopharm Chemical Reagent Co., Ltd. (Beijing, China).

All other chemicals and solvents were analytical grade.
