Isolation and Characterization of Chestnut Lipase

Korean chestnuts (Castanea crenata) were purchased from local markets. They were peeled, sealed in a vacuum plastic bag, and stored at 4 °C for subsequent use. Before use, the chestnuts were covered with a layer of wet cotton and allowed to germinate for 3 days at 18 °C.
HiTrap DEAE Sepharose Fast Flow, HiTrap Q Sepharose Fast Flow, and HiPrep Sephacryl S-100 HR were purchased from Cytiva (Uppsala, Sweden). Trizma® base (≥ 99.9%), diethyl ether (≥ 99.9%), p-nitrophenyl palmitate (p-NPP), sodium dodecyl sulfate (SDS), isooctane (IOT), tributyrin, tricaproin, tricaprin, trilaurin, tripalmitin, and triolein (≥ 99.0%) were purchased from Sigma-Aldrich Co. (St. Louis, MO, USA). Ammonium sulfate (≥ 99.5%), Triton X-100, sodium chloride (≥ 99.5%), and agar were purchased from Junsei Chemical Co., Ltd. (Tokyo, Japan). Boric acid, potassium chloride, and oleic acid were obtained from Duksan Pure Chemicals Co., Ltd. (Ansan, Gyeonggi-do, Korea). The Costar® 96-well microplates (clear wall, clear bottom) used for the fluorometric assay were purchased from Corning Co. (Corning, NY, USA). All other chemicals were of analytical grade and were used without further purification.

Lipid Extraction and Analysis Protocol

Synthesis and Characterization of Dicaprin Stereoisomers

1,2-sn- and 1,3-sn-dicaprin were from Sigma. 2,3-sn-dicaprin was prepared from tricaprin (Sigma) by performing stereospecific enzymatic hydrolysis on the sn-1 ester bond [17] (link). As expected, the surface pressure-molecular area curves obtained with 1,2-sn-and 2,3-sn-dicaprin were found to be superimposable and characteristic of the 1iquid expanded state, and showed no sign of discontinuity throughout the range of surface pressures investigated. The collapse pressure of 1,2-sn- and 2,3-sn-dicaprin was 40 mN.m−1, and that of 1,3-sn-dicaprin was 32 mN.m −1[18] .

Sterol standards, tricaprin, glycerin, monoolein, diolein, triolein, butanetriol, and betulin were purchased from Sigma-Aldrich (St. Louis, MO, USA) and Acros Organics (Morris Plains, NJ, USA). All reagents used in this study were purchased from Sigma-Aldrich, Merck (Darmstadt, Germany), Chem-Lab (Zedelgem, Belgium), or Fisher Scientific (Hampton, NH, USA) and were either analytical or HPLC grade.

Glyceride Standards for Analytical Methods

Stock solutions of glycerides were prepared in dichloromethane. A 1 μM glyceride mixture in 2 : 1 : 1 isopropanol/ acetonitrile/water was used as a quality control standard. All standard solutions were stored at -24 °C before and after analysis to prevent degradation and solvent evaporation.

Enzymatic Production of Cocoa Soapstock Lipids

The raw material used in the experiments was an olein from cocoa (Theobroma cocoa) soapstocks, obtained from OLEOFAT TRADER S.L. (Tudela -Navarra -Spain) by acid fat-splitting. Cocoa soapstocks were provided by Moner Cocoa S.A. (La Selva del Camp -Tarragona -Spain). The enzymes Lipozyme CALB and Eversa transform 2.0 were purchased from Novozymes (Bagsvaerd -Copenhagen). Glycerol (99.9%) was purchased from Labbox, NaOH, KOH and the solvents used for analysis (ethanol, n-hexane, diethyl ether, methanol and acetone) were from Scharlab. The HMDS+TMCS+Pyridine 3:1:9 (Sylon HTP) mix used to quantify MAG, DAG and TAG and the internal standards such as 1,2,4-butanetriol and tricaprin were purchased from Supelco (Sigma Chemical Co., St Louis, MO). The standards for the calibration curves in the GC analysis, 1-Monolarin, Diolein, Triolein and Glycerol, were purchased from Larodan and the 30 mg/ml FAME mix (28 components) was purchased from Restek.
