Quantification of Caspase-3 Activity

Caspase-3 Expression in Core-Shell Treated Cells

The cells were cultured in RPMI medium in a 6 well plate at a density of 3 x 105 cells/2 mL and incubated in a CO2 incubator at 37°C for 24h. The spent medium was then aspirated and the cells were treated with the core-shells dispersed in fresh medium and retrieved for caspase-3 expression accordingly for four time intervals (2, 6, 12, 24h). At each time interval, the medium was removed from the wells, washed with PBS and trypsinised. Harvested cells were centrifuged at 2000 rpm and supernatant was decanted. Caspase expression was estimated using FITC Rabbit Anti- Active Caspase– 3 (BD Biosciences, Catalog no.560901). Further analysis was done in flow cytometry (BD FACS Caliber).
