Chronic Implantation of Electrodes for EEG and EMG Recording

All surgical procedures were performed stereotaxically under aseptic conditions, as described previously [31 (link), 44 (link)]. Briefly, while animals were under isoflurane anesthesia (5% induction; 2%–3% maintenance in 100% oxygen), electrodes were chronically implanted to enable recording of cortical electroencephalogram (EEG), hippocampal EEG, and nuchal electromyogram (EMG). All electrodes were secured to the skull with dental acrylic, crimped to miniconnector pins, and brought together in a plastic connector, which was secured to the skull with additional dental acrylic. Upon completion of the surgical procedure, animals were administered saline (5 cc, s.c.) to prevent dehydration. Postoperative pain was controlled with buprenorphine (0.05 mg/kg, I.M.; Cerilliant, Round Rock, TX, USA).

Opioid Receptor Binding Assay

Morphine, hydromorphone, oxymorphone, butorphanol, levorphanol, nalbuphine, nalorphine, naltrexone, 6β-naltrexol, buprenorphine, pentazocine, and naloxone, were purchased from Cerilliant. HepG2 human hepatoma cell line (ATCC HB-8065) was obtained from American Type Culture Collection. Pooled human lung S9 fraction, liver cytosol, small intestine (duodenum and jejunum) S9 fraction, and kidney S9 fraction were purchased from XenoTech, LLC. All other chemicals were of the highest grade commercially available.

Quantitative Analysis of Opioids and Antipsychotics

Surine DOA (synthetic urine) was purchased from Dyna-Tek Industries (Lenexa, KS). Buprenorphine, norBuprenorphine, naloxone and their respective deuterated internal standards were obtained from Cerilliant (Austin, TX). For quetiapine metabolite characterization experiments, quetiapine, 7-hydroxyquetiapine and quetiapine acid were also acquired from Cerilliant, and quetiapine sulfoxide was purchased from Toronto Research Chemicals (Toronto, Ontario, Canada). HPLC-grade acetonitrile, methanol and ammonium hydroxide (ACS grade) were obtained from Thermo Fisher (Waltham, MA). Formic acid (98%, ACS grade) was purchased from EMD Millipore (Billerica, MA), and phosphoric acid (ACS grade) was purchased from Sigma-Aldrich (St. Louis, MO). Purified water was supplied by an in-house MilliQ system (EMD Millipore). IMCSzyme recombinant β-glucuronidase and Rapid Hydrolysis Buffer (pH 7.2–7.8), used for enzymatic hydrolysis in all laboratories, were purchased from IMCS (Integrated Micro-Chromatography Systems, Irmo, SC).

Buprenorphine-loaded PLGA Nanoparticles
