Fluorescence Imaging of Live Parasites

Check if the same lab product or an alternative is used in the 5 most similar protocols
Fluorescence images were taken of live parasites on a Leica D6B fluorescence microscope equipped with a Leica DFC9000 GT camera and a Leica Plan Apochromat 60× or 100× / 1.4 oil objective. Contrast and intensities were linear adjusted to present clear parasite shapes using Fiji and cropped images were assembled into a composition of cells for Fig. 1 using Adobe Illustrator CC 2021.

Quantitative Analysis of Fluo-CQ Uptake

Check if the same lab product or an alternative is used in the 5 most similar protocols
Giemsa-stained culture smears were imaged on a DM2000 LED light microscope (Leica) equipped with a DMC2900 camera (Leica). Fluorescence microscopy was performed with an Eclipse Ni light microscope (Nikon) fitted with a C11440 camera (Hamamatsu) or with a D6B fluorescence microscope (Leica) equipped with a DFC9000 GT camera (Leica). For detailed microscopy protocols including quantitative analysis of Fluo-CQ uptake, please refer to SI Appendix.

Live-cell Nuclei Staining Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
For live-cell microscopy, LCN-Kd parasites were incubated with 1 mg/ml DAPI in culture medium for 15 min at 37 C to stain nuclei. Parasites were then washed once in PBS and imaged in PBS on a Leica D6B fluorescence microscope equipped with a Leica DFC9000 GT camera and a Leica Plan Apochromat 100x/1.4 oil objective. Image processing was performed using ImageJ (Schneider et al., 2012) .

Oxidative Stress Measurement in Malaria Parasites

Check if the same lab product or an alternative is used in the 5 most similar protocols
For measurement of oxidative stress levels, LCN-Kd and LCN-Con ring stage parasites were treated with 2.5 mM glucosamine or were left untreated. At around 30 hpi, trophozoite stage parasites were stained with 5 mM CellROX Orange (Thermo Fisher Scientific) in culture medium for 30 min at 37 C. After three washes with PBS, they were directly imaged on a Leica D6B fluorescence microscope as described above. Mean fluorescence intensities and size (area) of individual parasites were determined using the ROI manager of ImageJ (Schneider et al., 2012) .

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
