Polymorphprep and lymphoprep reagents

Manufactured by Axis-Shield
Sourced in Norway

Polymorphprep and lymphoprep reagents are density gradient media used for the separation and isolation of specific cell populations from whole blood. They facilitate the separation of mononuclear cells from erythrocytes and granulocytes through centrifugation.

Isolation of Healthy Neutrophils

Check if the same lab product or an alternative is used in the 5 most similar protocols
Human neutrophils were isolated from healthy blood donors using density gradient centrifugation of polymorphprep and lymphoprep reagents (AXIS-SHIELD PoC AS, Oslo, Norway) according to the manufacture’s instruction. The regional ethics review board in Uppsala, Sweden has given their approval (Dnr 2015/437) to isolate blood from healthy individuals after informed consent. The collection of blood from healthy donors were performed according to the ethical guidelines of both the Declaration of Helsinki and the Swedish national board of health and welfare. The viability of neutrophils was examined by microscopy, as previously described22 (link).

Isolation of Healthy Human Neutrophils

Check if the same lab product or an alternative is used in the 5 most similar protocols
Human neutrophils were isolated from healthy blood donors by density gradient centrifugation of Polymorphprep and Lymphoprep reagents (AXIS-SHIELD PoC AS, Oslo, Norway) or with the EasySep Human Neutrophil Enrichment Kit (STEMCELL Technologies, Cambridge, UK) according to the manufacturer’s instructions32 (link). The cells were infected in RPMI 1640 (Lonza, Basel, Switzerland) complemented with 10% heat inactivated fetal bovine serum (FBS) during the experiments. An ethical approval has been granted by the regional ethics review board in Uppsala, Sweden (Dnr 2015/437), to isolate blood from healthy individuals after informed consent. Blood from healthy donors were collected according to the ethical guidelines of both the Declaration of Helsinki and the Swedish national board of health and welfare. The viability of the neutrophils was > 90% as determined by the trypan blue exclusion test after isolation.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
