Quantifying Tumor IL-12 Levels

Tumors and serum were collected from animals at various time points after a single treatment. The tumors were mechanically homogenized in cold phosphate buffered saline (Mediatech, Manassas, VA) containing protease inhibitor (Roche, Indianapolis, IN) using a tissue homogenizer (Omni International, Kennesaw, GA). The homogenate was centrifuged and the supernatant collected and assayed for IL-12 expression using a mouse IL-12 ELISA kit (R&D Systems, Minneapolis, MN) according to the manufacturer’s instructions.

Cytokine Quantification in Tumor Tissues

For in vivo assays, tumor tissues were minced, homogenized, and subjected to beads heating in PBS with protease inhibitor cocktail (K1007, APExBIO) on day 3 or day 6 after completion of treatment. For in vitro tests, culture media were collected by centrifugation. Quantification of cytokines in tumors and media was performed by Mouse IL-12 ELISA Kit (M1270, R&D) and Mouse IFN-γ ELISA Kit (E-EL-M0048c, Elabscience) following the manufacturer’s instruction, respectively.

Assessing IL-12 Levels in Mouse Model

A single injection of 50 μg of scIL-12/CMVpDNA intramuscularly or a single intraperitoneal injection of 100 ng recombinant mIL-12 was given to five LSL-Braf V600E /TPO-Cre mice, respectively. Serum was collected either every 3 days for 2 weeks following scIL-12/CMVpDNA injection or every 2 for 10 h after recombinant mIL-12 injection. Serum IL-12 levels were analyzed using a mouse IL-12 ELISA Kit (R&D Systems, Minneapolis, MN, USA).
