Fabrication of PU-Amla Oil Nanofibers

The homogeneous solution of Polyurethane (Tecoflex EG-80A, Lubrizol) and amla oil (obtained locally) were prepared by dissolving in Dimethylformamide (DMF) (Sigma Aldrich, UK) to prepare the homogeneous solution. In this present study, PU (9 wt%) solution was prepared and blended with 4 wt% concentration of amla oil to fabricate the electrospun nanocomposite membrane. The amla oil homogeneous solution was mixed to PU homogeneous solution at a ratio of 8:1 (v/v). The PU and PU amla oil membranes were fabricated through conventional electrospinning setup. The prepared homogeneous solutions were taken in the 10 ml syringe and attached to the syringe pump. For both samples, the nanofibers were attained at a voltage power supply of 10 kV was with a flow rate of 0.5 ml/hr. The nanofibers were collected on the collector drum which was placed 20 cm from the needle. Finally, the fabricated nanofibrous scaffolds were vacuum dried for 24 h to remove the residual DMF.

Biocompatible Polyurethane Membrane Synthesis

Tecoflex EG-80A (medical grade polyurethane) was supplied from Lubrizol and dissolved in dimethylformamide (DMF) solvent (Sigma Aldrich, Dorset, UK). Cytotoxicity Evaluation of MWCNT-Embedded Tecoflex®

Tecoflex® EG-80A was kindly supplied by Lubrizol Advanced Materials. Tetrahydrofuran (THF, 99+%) and N,N-dimethylformamide (DMF, 99.8%) were purchased from Sigma-Aldrich and used without further purification. MWCNTs (Baytubes® C 150 HP outer diameter of ~ 13 nm, inner diameter of ~ 4 nm and length of >1 μm) were kindly supplied by Bayer Materials. Mouse embryonic NIH 3T3 fibroblasts were purchased from ATCC (Manassas, VA). Dulbecco’s Modified Eagle Medium (DMEM) supplemented with 10% newborn calf serum (NBCS), 7.5% sodium bicarbonate, and 1% antibiotic-antimycotic 100× (10000 units/mL of penicillin, 10000 μg/mL of streptomycin, and 25 μg/mL of amphotericin B) were obtained from Invitrogen. 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT reagent) and Dymethyl sulfoxide (DMSO, ≥99.9%) were acquired from Sigma-Aldrich, 0.4% Trypan blue and 0.25% Trypsin-EDTA for cell harvest from Invitrogen. Glycine was purchased from BioRad Laboratories to form Glycine buffer, which was prepared by taking 0.1M Glycine containing 0.1M NaCl, followed by equilibration with 0.1N NaOH to pH = 10.5. Treated tissue culture flasks and microplates for cell growth and seeding were purchased from Fisher Scientific.

Thermoplastic Polyurethane Fabrication and Characterization

Fabrication of PU-Gelatin Electrospun Scaffolds

The scaffolds were produced using an NF-103 (MESS, Fukuoka, Japan) electrospinning device under the conditions previously described [24 (link)]. PU-gelatin scaffolds were made at a collector rotation speed of 300 rpm, a voltage of 18.5–19 kV, and a solution flow rate of 1.1–1.2 mL/h. The electrospinning solution was composed of 3% Tecoflex EG-80A polyurethane (batch 01016300734, Lubrizol Advanced Materials, Beveren, Belgium) (w/v) and 15% gelatin from porcine skin (Sigma № G2500, Saint Louis, MI, USA) (w (gelatin)/w (PU)) in 1,1,1,3,3,3-hexafluoroisopropanol (Sigma, Saint Louis, MI, USA). After preparation, scaffolds were dried in a fore vacuum for at least 3 h to remove residual solvent before being stored in sealed zip-lock bags.
Flat films (100–120 µm thick) were obtained by casting a glass slide with 3% Tecoflex EG-80A polyurethane solution (w/v) in 1,1,1,3,3,3-hexafluoroisopropanol at room temperature (23–25 °C). After 3 days of incubation, fabricated flat films at room temperature were additionally dried using a fore vacuum to remove the residual solvent and stored in sealed zip-lock containers.
