Immunohistochemical Analysis of Thyroid Lesions

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immunohistochemistry (IHC) analysis was performed to study altered protein expression in formalin-fixed and paraffin-embedded human thyroid lesions. The following primary antibodies were used: anti-Trop2 (R&D Systems, Minneapolis, MN), and anti-MMP2 (Lifespan Bioscience, Seattle). The degree of immunostaining was examined and scored independently by two observers by combining both the proportion of positively staining tumor cells and the staining intensity as previously described [23 (link)].

Quantifying Protein Expression via Western Blotting

Check if the same lab product or an alternative is used in the 5 most similar protocols
Western blotting (WB) was performed according to a standard method as described previously [25 (link)]. The following primary antibodies were used: anti-Trop2 (R&D Systems, Minneapolis, MN), anti-ERK1/2 and anti-phospho-ERK1/2 (Cell Signaling Technology, Beverly, MA), anti-p38, anti-phospho-p38, anti-JNK, and anti-phospho-JNK (Abcam, Cambridge, MA), and anti-α-tubulin (Sigma-Aldrich, St. Louis, MO).

Trop2 Immunohistochemistry Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immunohistochemistry was conducted on a 4-μm tissue array section using a BOND-MAX automated immunostainer and a Bond Polymer Refine Detection kit (Leica Microsystems, Wetzlar, Germany) according to the manufacturer’s instructions. The primary antibody used was anti-Trop2 (R&D Systems, Minneapolis, MN, USA; 1:800). The expression of Trop2 was determined by examining the cell membrane and the intensity and percentage of cells expressing Trop2 were evaluated. Histology index (H index) was calculated by multiplying the intensity score (0 = negative; 1 = weak; 2 = moderate; 3 = strong) and percentage of positive cells (range = 0–100), to obtain a score range from 0 to 300.

Pyrrole, poly(sodium 4-styrenesulfonate) (PSS), N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride (EDC), N-hydroxysuccinimide (NHS), streptavidin, and sodium hydroxide (NaOH) were obtained from Sigma Aldrich (St. Louis, MO, USA). Anodized aluminum oxide (AAO) membrane filter (pore diameter, 200 nm) was purchased from Whatman (Pittsburgh, PA, USA). Silver paste (P-100 type) was obtained from CANS. NHS-SS-biotin was purchased from CovaChem (Loves Park, Illinois, USA). Biotinylated anti-EpCAM, anti-EGFR, anti-vimentin, anti-N-cadherin, and anti-Trop2 were obtained from R&D system (Minneapolis, MN, USA). Biotinylated anti-CD63 and anti-CD81 were purchased from AnCell (Oak Park, Minnesota, USA). Biotinylated anti-CD9 was obtained from Abcam (Cambridge, UK).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
