Antimicrobial Susceptibility of Bacterial Pathogens

Staphylococcus aureus GRP-0057, Klebsiella pneumoniae GRP-0107, and Pseudomonas aeruginosa strains GRP-0019, GRP-0049, GRP-0036, and ATCC 27853 were used to infect the mice in the different models. The microorganisms were grown to log phase in Mueller-Hinton broth and agar for susceptibility tests, and in trypticase soy broth and agar for animal infection models (all from Becton Dickinson, Sparks, MD, USA). Staphylococcus aureus ATCC 29213, K. pneumoniae ATCC 43816 and P. aeruginosa 27853 were the quality control strains in susceptibility tests; Kocuria rhizophila ATCC 9341 was the seeding organism for microbiological assays. We bought the antibiotics from well-reputed local drugstores and reconstituted them following manufacturer instructions. The products, both licensed for human use by INVIMA (the Colombian DRA), included a generic product of imipenem (Inem, Ivax Pharmaceuticals, Mexico) and the innovator (Tienam, Merck Sharp & Dohme, Elkton, VA). Both products came in vials with a lyophilized powder containing imipenem 500 mg and cilastatin 500 mg; lot numbers and additional information about the products employed in this study are described in S1 Table.

Cucurbit[n]urils as Potential Therapeutics

Cucurbit[n]urils (n = 6, 7 and 8) were synthesized, purified and kindly provided by the Nikolaev Institute of Inorganic Chemistry (Novosibirsk, Russia). Before the experiments, CB[n] was diluted in the culture medium RPMI-1640. CB[6] and CB[7] were prepared at a concentration 0.3, 0.5 and 1 mM; CB[8] at 0.01 mM and in a PBS solution; CB[6] and CB[7] at 0.2, 0.5, 1 and 2 mM; CB[8] at 0.01 mM. RPMI-1640 (Biowest LLC, Riverside, MO, USA), gentamicin (Dalfarma, Khabarovsk, Russia), tienam (Merck Sharp & Dohme Corp., Kenilworth, NJ, USA), FCS (Hyclone, Chicago, IL, USA), tabs for preparing phosphate-buffered saline (PBS) (AppliChem GmbH, Darmstadt, Germany), Na2EDTA (Helicon, Moscow, Russia), Ficoll (BioClot, Aidenbach, Germany), urografin 76% (Schering, Berlin, Germany), human albumin solution (Microgen, Moscow, Russia), Triton X-100 (Dow Corning, Midland, MI, USA, WST-1 assay kit (Takara Bio, Kusatsu, Japan), LDH-Cytox™ Assay Kit (BioLegend, San Diego, CA, USA), PE Annexin V Apoptosis Detection Kit with 7ADD (BioLegend, San Diego, CA, USA), anti-CD3 antibody (BioLegend, San Diego, CA, USA) and anti-CD4 antibody (BioLegend, San Diego, CA, USA).
The Ethical Committee of RIFCI, Russia, approved the study design and the recruitment of subjects. Subjects provided written informed consent. The relevant guidelines and regulations were followed when performing the experiments.

Cell Culture of HTLV-1 and CML Lines

Human T-cell leukemia cell line 1301 and human chronic myelogenous leukemia cell line K562 (European collection of authenticated cell cultures, Sigma Aldrich, Merck KGaA, Germany) were used. Cells were cultivated in the RPMI-1640 cell medium containing 10% foetal calf serum (HyClone Laboratories, South Logan, UT, USA), 0.3 mg/mL L-glutamine (Vector, Russia), 50 µg/mL gentamicin (DalChimPharm, Khabarovsk, Russia), and 25 µg/mL tienam (Merck Sharp & Dohme, Kenilworth, NJ, USA) in a humidified atmosphere containing 5% CO2 at 37 °C.

Simvastatin and Imipenem-Cilastatin Sodium Protocol

Simvastatin (SV) was purchased from MSD Pharmaceutical Company Limited (Hangzhou, China). TIENAM (Imipenem:Cilastatin Sodium=1:1) was obtained from Merck Sharp & Dohme Corp. All the primary and secondary antibodies for the western blot analysis were purchased from Cell Signaling Technology (USA). TRIzol reagent used for total RNA extraction was purchased from Sigma-Aldrich (USA). High capacity cDNA reverse transcription kit and SYBR Master Mix were purchased from Applied Biosystems (USA); RIPA Lysis Buffer was purchased from Beyotime company (China). SuperSignal West Pico Chemiluminescent Subatrate was purchased from Thermo Fisher Scientific (USA). The derivatization reagent for GC-MS analysis, N,O-Bis(trimethylsilyl)trifluoroacetamide (BSTFA) with 1% trimethylchlorosilane (TMCS) was purchased from Regis (Morton Grove,IL, USA).
