Human skeletal cells were separated from red blood cells (RBCs) and bone dust by density gradient separation using 1:1 Histopaque of density 1.119 g/mL (Sigma-Aldrich, St Louis, MO, USA). The buffy coat was collected, washed with staining media, and the resulting cell suspension was depleted of CD45 + cells by magnetic-activated cell sorting (MACS) (Miltenyi, Cat#130–045-801).
