Socs3 sirna

Manufactured by Thermo Fisher Scientific
Sourced in United States

SOCS3 siRNA is a small interfering RNA (siRNA) molecule designed to specifically target and downregulate the expression of the SOCS3 gene. SOCS3 is a negative regulator of cytokine signaling pathways. The SOCS3 siRNA can be used as a research tool to investigate the role of SOCS3 in various cellular processes.

Intracerebroventricular Injection Procedure for siRNA and CRISPR Delivery

Check if the same lab product or an alternative is used in the 5 most similar protocols
Intracerebroventricular injection (i.c.v) method was performed as previously reported.14 (link) Briefly, the rats were placed in a stereotaxic apparatus under 3% isoflurane anesthesia. A burr hole was drilled into the skull at the following coordinates relative to the bregma:1.5mm lateral, 0.9mm posterior, and the 10-μL Hamilton syringe was inserted into the burr hole at a depth of 3.3mm. SOCS3 siRNA (Ambion, USA) and control siRNA were infused into the right ventricle at a rate of 1 μL/min with a pump. H3K27me3 CRISPR (Santa Cruz Biotechnology, Dallas, TX, USA) was used to increase H3K27me3 gene expression. A total of 5ug per pup of H3K27me3 CRISPR or scrambled CRISPR (Santa Cruz Biotechnology, Dallas, TX, USA) was injected into the ipsilateral ventricle at 48h before SAH.

Neuroprotective Effects of EPZ6438 in Rat SAH Model

Check if the same lab product or an alternative is used in the 5 most similar protocols
EPZ6438 (TargetMol, USA) was dissolved in 10% DMSO and administered via intraperitoneal injection (i.p). Three different dosages of EPZ6438 (1mg/kg, 3mg/kg, 9mg/kg) were given to rats at 1h after SAH.7 (link) SOCS3 siRNA (Ambion, USA) was administrated intracerebroventricularly. The same volume of 10% DMSO was given via i.p as the vehicle.

Modulation of SOCS3 Expression

Check if the same lab product or an alternative is used in the 5 most similar protocols
Hsa-miR-455-5p mimics and a hsa-miR-455-5p inhibitor were purchased from Sigma-Aldrich. SOCS3 siRNA was obtained from Life Technologies. H460 and H1299 cells were transfected using X-tremeGENE (Roche Applied Science), and HEK293T cells were transfected using Lipofectamine 2000 (Invitrogen) according to the manufacturer’s directions. SOCS3 siRNA was transfected into cells for 24 hours using Lipofectamine RNAiMAX Reagent (Invitrogen).

SOCS3 Knockdown in PBMCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Isolated PBMCs were cultured as mentioned previously. Plated cells were transfected with siRNAs using linear polyethylenimines (PEIs) Reagent (Sigma-Aldrich, St. Louis, MO, USA) in accordance with the manufacturer’s instructions [11 (link)] with either SOCS3-siRNA (ID: s17189, Invitrogen) or scrambled (control) siRNA (cat. no. 4390844, Invitrogen). Gene knockdown efficiency was examined by qRT-PCR.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
