Comprehensive Clinical Monitoring of Macaques

Macaques were monitored for clinical signs at least twice daily throughout the experiment using a standardized scoring sheet as previously described (13 (link)). On exam days (Fig. 1a), clinical parameters such as body weight, body temperature, and respiration rate were collected, as well as ventrodorsal and lateral chest radiographs. Chest radiographs were interpreted in a blind manner by a board-certified clinical veterinarian. The total white blood cell, lymphocyte, neutrophil, platelet, reticulocyte and red blood cell counts and hemoglobin and hematocrit values were determined from EDTA-treated blood using an IDEXX ProCyte DX Analyzer (IDEXX Laboratories). Necropsies were performed after euthanasia, and gross pathology was scored in a blind manner by a board-certified veterinary pathologist. Histopathological analysis of tissue slides was performed by a board-certified veterinary pathologist who was blind to the group assignment of the macaques.

Comprehensive Blood Analysis in CKD Mice

Blood was collected via retro-orbital puncture 6 weeks through the experimental period and from the inferior vena cava at euthanasia. Blood urea nitrogen (BUN) and creatinine were measured on the Beckman Coulter (Brea, CA) AU 680 analyzer. Serum iron was measured using colorimetric assay (Pointe Scientific, Canton, MI). Genetic Models for Platelet Research

Age-matched mice were used wherever applicable and sexes were mixed for all experiments unless otherwise mentioned. Blood was drawn via cardiac puncture and whole blood counting was done with a Hemavet (Drew Scientific, Dallas, TX) and IDEXX ProCyte Dx analyzer (IDEXX Laboratories, Westbrook, ME). For assays, platelet counts were measured using the Z2 Coulter Counter (Beckman Coulter, Inc., Miami, FL).
