Quantification of Cyclosporine A Using CMIA

CsA (99% purity) was purchased from The European Directorate for the Quality of Medicines & Healthcare and CsA-d4 (95% purity) was purchased from Toronto Research Chemicals. Chromatographic grade acetonitrile, methanol, ammonium acetate and formic acid, were obtained from Thermo Fisher Scientific, Inc. Zinc sulfate heptahydrate (analytical grade) was obtained from Sinopharm Chemical Reagent Co., Ltd. Deionized water was purified with a Milli-Q Plus Ultrapure water system (Millipore Corp.). The CMIA method was performed on an Abbott ARCHITECT i1000SR system (Abbott Laboratories) and required a calibrator and CsA kit (cat. no. 1L75-25); these were also obtained from Abbott Laboratories. The acceptable ranges of CMIA quality control samples were defined as per the manufacturer's instructions.

Heavy Metal Toxicity Evaluation Protocol

Sprague Dawley (SD) rats were obtained from the Zhejiang Provincial Laboratory Animal Science Center (Hangzhou, Zhejiang, People's Republic of China). Water maze testing was performed using a Morris water maze video analysis system (RD1101-MWM-G, Shanghai Mobiledatum Information Technology Co., Ltd, People's Republic of China).

VAMS-Based Bioanalytical Assay

VAMS samplers (MitraTM, 20 μL microsampling device) were supplied by Neoteryx (Torrance, CA, USA). HPLC-grade methanol, acetonitrile, and formic acid were obtained from Thermo Fisher (St. Louis, MO, USA). Ammonium acetate was obtained from Sigma-Aldrich (St. Louis, MO, USA). Zinc sulfate heptahydrate was obtained from Sinopharm Reagent (Shanghai, China). Deionized water was prepared by a Milli-Q integral ultrapure water machine (Merck Millipore, Germany).

Pig Farm Wastewater Treatment Protocol

(Beijing, China). All the materials were used without further purification.
Acetate–sodium acetate buffer was prepared with 25 g/L sodium acetate and 10 mL/L acetic acid. The pH was adjusted to 4.8~5.0. For Fenton’s reagent, 1.52 mL of 8% ferrous sulfate was mixed with 0.2 mL of 30% hydrogen peroxide in 100 mL of distilled water, and the pH was adjusted to 2–4 with hydrochloric acid.
Pig farm wastewater samples were collected from a large-scale pig farm located in Huaian, Jiangsu Province, China, in June 2023. Wastewater is produced during the pig breeding process. The raw wastewater samples were collected before they were discharged into the wastewater treatment plant.
