Riboflavin Production Quantification

The amount of riboflavin produced was estimated after 48 h of fermentation. The fermented culture broth was centrifuged at 4800 g for 10 min. The collected supernatant was analyzed for the presence of riboflavin by using UV spectrophotometer at 444 nm. The concentration of riboflavin was calculated using extinction coefficient (1.04 × 10–2 M–1 cm–1) and the standard graph was plotted using riboflavin standard (Sigma Aldrich) (Sauer et al., 1996 (link)).

Determination of Riboflavin in Nutritional Formulas

Solvents were of "gradient grade for HPLC" and other chemical products were of "analytical grade" and all were purchased from Sigma-Aldrich (St Louis, USA) or VWR (Radnor, USA). For enzymatic hydrolysis, papain from Papaya latex and acid phosphatase from potato were purchased from Sigma-Aldrich.
