Expression of CD4, CXCR4, and CD3 on CD4+ T cells was analyzed by flow cytometry. Cells were stained for 15 minutes at room temperature in phosphate-buffered saline (PBS) containing 2% bovine serum albumin (BSA) with the following antibodies: Pacific Blue–conjugated anti-CD3 (Becton Dickinson), APC-Cy7-conjugated anti-CD4 (Becton Dickinson), and PE-conjugated anti-CXCR4 (Becton Dickinson), all of which were performed following the vendor’s protocol (Abcam). The cell preparations were analyzed with a FACSCalibur flow cytometer (Becton Dickinson). Positive and negative events were determined with matching isotype controls.
