Neutrophil and Renal Biopsy Staining

Check if the same lab product or an alternative is used in the 5 most similar protocols
Neutrophil samples were stained with anti-MPO-FITC antibody and mounted in DAPI-containing ProLong Gold Antifade Mountant (Thermo Fisher Scientific). AIM2 and IFI16 proteins were expressed, purified, and fluorescently labeled as previously described (Morrone et al., 2014 (link); Morrone et al., 2015 (link)). SLE renal biopsies were stained as previously described (Antiochos et al., 2018 (link)) using anti-MPO rabbit polyclonal (ThermoFisher), anti-MPO mouse monoclonal (ThermoFisher), anti-IFI16 mouse monoclonal (Sigma), anti-AIM2 rabbit polyclonal (Sigma), and Hoechst 33,342 (ThermoFisher). Confocal imaging was performed with a Zeiss AxioObserver with 780-Quasar confocal module.

Fluorescence Correlation Spectroscopy of mCherry-myoII in Dictyostelium

Check if the same lab product or an alternative is used in the 5 most similar protocols
Dictyostelium cells transformed with mCherry-myoII were plated on glass dishes. FCS experiments were performed on interphase cells at ambient temperature (∼20–23°C; normal growth temperatures for Dictyostelium cells) using a Zeiss AxioObserver with 780-Quasar confocal module and FCS, with a C-Apochromat 40× (NA 1.2) water objective. 100 nM Rhodamine 6G was used for pinhole alignment and structural parameter calculation. A structural parameter of six was used, and the confocal volume was measured to be 0.45 fL. Data were fit to a single-component 3D diffusion model, including a triplet-state component with an upper limit of 8 µs (Srivastava and Robinson, 2015 (link); Kothari et al., 2017 (link)).

Multi-Modal Confocal Microscopy Protocols

Check if the same lab product or an alternative is used in the 5 most similar protocols
Confocal microscopy was performed on a Zeiss AxioObserver with 780-Quasar confocal module & FCS (NIH Grant S10 OD016374), a Zeiss AxioObserver with LSM700 confocal module (NIH Grant S10 OD016374), or a Zeiss Axiovert 200 with 510-Meta confocal module. All DIC microscopy was performed with a Zeiss Axiophot 2 with Janoptik ProgResC14 Plus opitical imaging system.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
