Isolation and Characterization of Mesenchymal Stem Cells

Matrigel were obtained from Corning. Trizol Reagent were purchased by Invitrogen. All-In-One RT MasterMix Kit, EvaGreen qPCR MasterMix Kit were obtained from abmGood. Cell-tracker dye CM-DiI were purchased from Invitrogen.

EV Uptake Measurement in Cells

EV suspensions were incubated with 2 μM CellTracker dye CM-DiI (Invitrogen) in Hank’s balanced salt solution for 5 min, at 37°C, and subsequently placed on ice for an additional 15 min. Excess dye was removed using an exosome spin column (MW 100,000), as per the manufacturer’s instructions. For uptake analysis, stained EVs from 0.5 ml of medium (approximately 0.5 × 109 EVs) were cultured with HUVECs and PBMCs for 2 hours. Cells were then rinsed twice with PBS, and then fixed with 4% paraformaldehyde. Immunofluorescence images were captured with a confocal fluorescence microscope (Nikon A1R).
