Cell Lysis and Protein Extraction

Cells were washed with PBS and harvested with RIPA lysis buffer (50 mM Tris (pH 7.5), 150 mM NaCl, 1mM EDTA, 1% NP-40, 0.1% SDS) supplemented with protease and phosphatase inhibitors (1 mM PMSF, 10 μg/mL Leupeptin, 50 μg/mL TLCK, 50 μg/mL TPCK, 1 μg/mL Aprotinin, 1 mM NaF, 5 mM NaPPi, and 10 mM Na3VO4).
