Determination of cytokines in culture supernatants was performed with ELISA kits for TNF-α and Lcn2 (from R&D; Wiesbaden, Germany) and IL-6, IL-10 (from BD), following manufacturer’s instructions. In brief, CellROX®DeepRed was added at the concentration of 500 nM to the infected or induced cells and incubated for 30 min at 37°C. SYTOX Blue Dead Cell stain (1 mM) was then added and the cells analyzed by FACS sorting within the next 120 min. The following controls were included: Cells treated with N-acetyl-L-cysteine (NAC) or tert-butyl hydroperoxide (TBHP), and an unstained cell sample.
