Protein Extraction and Western Blot Analysis

For protein extraction, cells were washed with PBS and lysed using RIPA buffer (50 mM Tris-Cl, pH 7.4, 150 mM NaCl, 1% (w/v) sodium deoxycholate, 0.1% (v/v) SDS, 1% (v/v) Triton X-100, 2 mM EGTA, 2 mM EDTA, 50 mM NaF, 5 mM Na₂P₂O₇, 1 X complete protease inhibitor cocktail (Roche) and 0.1 M Sodium Orthovanadate). For PARP cleavage analysis, dead cells were collected by centrifugation and combined with live cell lysates. Lysates were sonicated, centrifuged, and the supernatant quantified using the BCA Protein Assay Kit (Pierce, Thermo Scientific) according to the manufacturer’s instructions.
