Synaptic Protein Expression in Drosophila

Western blotting of whole adult head lysates (1 head/lane) was performed using standard laboratory procedures with anti-SYT1 (1:1,000, kindly provided by Noreen Reist) or anti-syntaxin (1:1,000, Developmental Studies Hybridoma Bank, Iowa City, IA). Visualization and quantification were done using a LI-COR Odyssey Imaging System (LI-COR Biosciences, Lincoln, MA). Immunostaining was performed on 3rd instar larvae at wandering stage larvae as described previously.9 (link) Rabbit myc antibody (1:1,000; Genetex) and anti-horseradish peroxidase (1:1,000; Jackson ImmunoResearch, West Grove, PA) were used for immunostaining. Confocal stacks of muscle 6 and 7 NMJs containing immunoreactive proteins were captured on a Zeiss Pascal Confocal with PASCAL software (Carl Zeiss MicroImaging, Inc.) using a 63 × numerical aperture 1.3 Plan Neofluar oil immersion lens (Carl Zeiss, Inc) and fluorescent secondary antibodies (Molecular Probes, Carlsbad, CA).
