All procedures in this study were approved by the Animal Care Committee of the University of Manitoba, which adheres to the principles for biomedical research involving animals developed by the Council for International Organizations of Medical Sciences. Male Sprague-Dawley rats were weaned at 3 weeks of age and randomly assigned to a LF diet (10% fat; Research Diets, D12450B) or HF diet (45% fat; Research Diets, D12451), both containing sucrose, for 12 weeks, as previously described [29 (link),65 (link)]. Metabolomics analysis was performed on a 1290 Infinity Agilent high-performance liquid chromatography (HPLC) system coupled to a 6538 UHD Agilent Accurate Q-TOF LC/MS equipped with a dual electrospray ionization source, as previously described [29 (link),65 (link)]. PCR-based gene expression arrays were purchased from SA Biosciences (Qiagen).

Lipidomic Analysis of Soleus Muscle

Check if the same lab product or an alternative is used in the 5 most similar protocols
Analysis of total soleus lipids was performed using a lipid soluble extraction, and analysis was preformed as described previously.5 (link) Briefly, lipid extracts were reconstituted with 100 μl of 80% acetonitrile prepared in deionized water. Metabolomics analysis was performed on a 1290 Infinity Agilent high-performance liquid chromatography (HPLC) system coupled to a 6538 UHD Agilent Accurate Q-TOF LC/MS equipped with a dual electrospray ionization source. A 3 × 50 mm, 2.7 μ Agilent Poroshell column was used to separate metabolites while the column temperature was maintained at 60 °C. The mass detection was operated using dual electrospray with reference ions of m/z 121.050873 and 922.009798 for positive mode; and m/z 119.03632 and 980.016375 for negative mode. The workflow utilized for data processing comprised several algorithms used by Agilent Mass Hunter Qualitative (MHQ, B.05, Agilent Technologies, Mississauga, ON, Canada) and by Mass Profiler Professional (MPP, 12.6, Agilent Technologies).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
