Lipid Extraction and Purification Protocol

All reagents and solvents were super purity or HPLC grade unless otherwise stated. Anhydrous pyridine, 2',7'-dichlorofluorescein solution, dichloromethane, and hexamethyl disilazane were from Honeywell (Fisher Scientific SL, Madrid, Spain). Acetone, ethanol, ethyl acetate, ethyl ether, methanol, n-hexane, n-heptane, propan-2-ol, propionitrile and trimethylchlorosilane were supplied by VWR International, LLC (West Chester, Pennsylvania, USA), anhydrous sodium sulfate and potassium hydroxide by Panreac Quimica, S.A.U. (Castellar del Valles, Barcelona, Spain). The standards 5α-cholestan-3ß-ol, α-, ß-, γ-, and δ-tocopherol, and n-eicosane (C20) were purchased at Sigma-Aldrich (Merck KGaA, Darmstadt, Germany). Silica gel 60 for column chromatography, 70-230 mesh (Merck KGaA, Darmstadt, Germany) was used directly from the container. Glass chromatography columns (50 cm length x 1.5 cm i.d.) were supplied with Teflon stopcocks; a plug of glass wool fiber was placed at the bottom and everything was washed with n-hexane before use.

Extraction and Derivatization of Analytes

Hydroxyl ammonium chloride, pyridine, methyl tertiary‐butyl ether, and sodium methylate for the extraction and derivatization were HPLC grade. N‐trimethylsilylimidazole and N‐Methyl‐N‐(trimethylsilyl) trifluoroacetamide were GC derivatization grade. Undecane, hexadecane, Tetracosane, triacontane, and octatriacontane were retention time standards; Tetracosane, 5α‐cholestan‐3ß‐ol, phenyl‐ß‐D‐glucopyranoside, and p‐chloro‐L‐phenylalanine were internal standards; and all reference standards were obtained from Sigma Aldrich Co. (St.Louis, MO, USA). Acetonitrile, hexane, methanol, and dichloromethane were analytical grade purchased from RCI Labscan Ltd. (Pathumwan, Bangkok, Thailand).

Cholesterol and Tocopherol Quantification

5-α-cholestan-3-ol and tocopherols (α, β, γ, δ) were standards (Purity 99%) supplied by Sigma-Aldrich Corp. (St. Louis, MO, USA). N,O-Bis(trimethylsilyl) trifluoro acetamide with 1% trimethyl chlorosilane was used as a silanization reagent and purchased from Supelco Inc. (Bellefonte, PA, USA). All other chemicals and reagents were of analytical grade and were acquired from local suppliers. Trisyl® (GRACE and Co., Columbia, MD, USA) and bleaching earths (Tonsil Optimum 278 FF and Tonsil Supreme 114 FF) were supplied by Clariant Iberica Prod. SA (Toledo, Spain).

Quantification of Lipid Standards

The 5-α-Cholestan-3-ol, heneicosanol, betulinic acid, squalane, and tocopherols (α, β, γ, δ) standards (Purity 99%) were supplied by Sigma-Aldrich SA (St. Louis, MO, USA). N,O-Bis (trimethylsilyl) trifluoroacetamide (BSTFA) containing 1% trimethylchlorosilane (TMCS) was purchased from Supelco (Bellefonte, PA, USA) and used as a silylation reagent. All other chemicals and reagents were of analytical grade and were acquired from local suppliers.
