Apoptosis Assay Using Flow Cytometry

Check if the same lab product or an alternative is used in the 5 most similar protocols
The apoptosis assay was carried out according to a previously described protocol (19 (link)). Briefly, the EC cells were transfected with siRNAs as previously described and incubated at 37°C for 96 h. The cells were harvested using trypsin and stained with the Fluorescein isothiocyanate (FITC) Annexin V Apoptosis Detection Kit II (BD Biosciences) at 25°C for 15 min. The cells were evaluated using FACS on a BD FACSCalibur HG Flow Cytometer Instrument (BD Biosciences) and Cell Quest Pro software version 3.1 (BD Biosciences), and the data were analyzed using FlowJo software version 16 (FlowJo LLC).

Cell Cycle Analysis of Transfected EC Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The cell cycle analysis was performed as previously described (19 (link)). In brief, EC cells were transfected with siRNAs as described earlier and incubated at 37°C for 96 h. The cells were harvested with trypsin and fixed with 70% ethanol at 4°C overnight and stained using propidium iodide (MilliporeSigma) at 4°C for 15 min. The cell cycle analysis was evaluated using fluorescence-activated cell sorting (FACS) on a BD FACSCalibur HG Flow Cytometer Instrument (BD Biosciences) and Cell Quest Pro software version 3.1 (BD Biosciences). The data were calculated using FlowJo software version 16 (FlowJo LLC).

Cell Cycle Analysis by FACS

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were fixed with 70% ethanol and incubated at 4 °C overnight. RNase A stock solution (final concentration: 0.5 mg/mL) was added and incubated for 20 min at 37 °C. Propidium iodide (PI, 50 mg/mL; Sigma-Aldrich; P4170) was added and incubated for 15 min at 4 °C in the dark. Cell cycle was measured by fluorescence-activated cell sorting (FACS) using a BD FACSCalibur™ HG Flow Cytometer Instrument (BD, Franklin Lakes, NJ, USA) and Cell Quest Pro software v3.1 (BD). FlowJo® v10 (BD Biosciences, San Jose, CA, USA) was used for the analysis. The experiment was repeated three times [24 (link)].

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
