Combinatorial Drug Synergy Evaluation

NVP-BEZ235 (S1009), cisplatin (S1166), 3-Methyladenine (S2767) and Chloroquine Phosphate (S4157) were obtained from Selleck Chemical in U.S.A. NVP-BEZ235 was provided as a 20 nM stock solution in 100% DMF. Final DMF concentration was kept constant at 0.1% in control and compound-treated cells. cisplatin solution was provided as a 1 mg/ml stock solution in 0.9% isotonic saline solution. 3-Methyladenine solution was provided as a 100 mM stock solution in PBS. Chloroquine Phosphatesolution was provided as a 50 mM stock solution in water. Working solutions were prepared freshly before addition to the cell media.N-Methylpyrrolidine (NMP,120-94-5) and Polyethylene glycol 300 (PEG300,1546423) were bought from Sigma-Aldrich.

Synthesis and Characterization of Zinc Electrolytes

ZnSO4·7H2O, ZnCl2, ZnBr2, ZnI2, Polyethylene glycol 300 (PEG300) and Deuterium oxide (D2O, ≥99.96%) were purchased from Sigma-Aldrich. In all, 0.25 mm Zn foil (99.994%) was bought from Alfa Aesar; 0.1 mm Zinc foil (99.999%), AC, DOL, LiNO3, LiTFSI and LiI were bought from Sigma. Chemicals are used as-is without any modifications. For 20-μm Zinc foil, 0.1 mm Zinc foil was rolled many times until reaching the desired thickness. Deionized water was obtained from Milli-Q water purification system. The resistivity of the deionized water is 18.2 MΩ cm−1 at room temperature. ZnSO4·7H2O, ZnCl2, ZnBr2 and ZnI2 were dissolved in deionized water to prepare 0.05 M, 0.5 M, 1 M electrolyte, respectively. Free-standing interwoven carbon fibers (Plain carbon cloth 1071 HCB) were purchased from Fuel Cell Store.

Synthesis and Characterization of Radiolabeled E2

Carprofen (Rimadyl) was purchased from Zoetis (Parsippany, NJ, USA). Tritiated E2, dissolved in ethanol, was purchased from American Radiolabeled Chemicals (St. Louis, MO, USA). Solvable Solution and Hionic-Fluor scintillation cocktail were purchased from PerkinElmer (Waltham, MA, USA). Randomly methylated β-CD, 2HP-β-CD, β-CD, γ-CD, polyethylene glycol 300 (PEG300), heparin sodium salt, and non-radioactive E2 were purchased from Sigma-Aldrich (St. Louis, MO, USA), and ethanol was purchased from VWR (Radnor, PA, USA). Additional chemicals and solutions, including sodium chloride, 30% hydrogen peroxide, monobasic sodium phosphate, and dibasic sodium phosphate, were purchased from Fisher Scientific (Waltham, MA, USA).

Rapamycin Dose Optimization for FaDu Xenografts

An initial dose-escalation study was performed to determine the optimal dose of rapamycin in FaDu xenograft-bearing BALB/c mice. FaDu xenografts were chosen as they have moderate VEGF production and relatively consistent tumour growth rates. The mice received 1, 5 or 20 mg/kg of rapamycin or vehicle via intraperitoneal injection daily for 10 days. The mice were assessed for systemic side effects of therapy, and tumour sizes were measured daily. Daily 20 mg/kg rapamycin appeared to have greater growth inhibition than the other doses and did not lead to significant systemic effects in the mice.
Subsequently, FaDu xenograft-bearing mice (n = 6–7/group) received rapamycin (20 mg/kg; LC laboratories) or vehicle (DMSO, 5% TWEEN-80 (Sigma-Aldrich), 5% polyethylene glycol 300 (PEG300) (Sigma-Aldrich) and 0.9% NaCl (Braun)) by daily intraperitoneal injection for 10 days. On day 5 of rapamycin therapy, the mice received 111In-bevacizumab (5 MBq, 3 μg) or 111In-IgG1κ (5 MBq, 3 μg) intravenously. After treatment, the mice were euthanised. Tumour size was measured with callipers, and volume was calculated using the formula: V = (a × b2)/2, where a and b are the largest and the smallest perpendicular diameters, respectively.

Synthesis of Sulfonated Polyamide Membranes

Only freshly sublimed terephthaloyl chloride and isophthaloyl chloride were utilized for the polymerization reactions. The diamine monomer, 4,4′-diaminobiphenyl-2,2′-disulfonic acid hydrate (95%) (BDSA) was purchased from Alfa Aesar and purified before use. Formulation and Evaluation of Piroxicam Delivery
