Rapamycin Dose Optimization for FaDu Xenografts
Subsequently, FaDu xenograft-bearing mice (n = 6–7/group) received rapamycin (20 mg/kg; LC laboratories) or vehicle (DMSO, 5% TWEEN-80 (Sigma-Aldrich), 5% polyethylene glycol 300 (PEG300) (Sigma-Aldrich) and 0.9% NaCl (Braun)) by daily intraperitoneal injection for 10 days. On day 5 of rapamycin therapy, the mice received 111In-bevacizumab (5 MBq, 3 μg) or 111In-IgG1κ (5 MBq, 3 μg) intravenously. After treatment, the mice were euthanised. Tumour size was measured with callipers, and volume was calculated using the formula: V = (a × b2)/2, where a and b are the largest and the smallest perpendicular diameters, respectively.

Polyvinyl alcohol 26-88 (PVA, Mw 160000) was obtained from Merck (Darmstadt, German) and polyvinylpyrrolidone 360 (PVP, Mw 360000) were purchased from Sigma-Aldrich (Schnelldorf, German). Poloxamer (Pluronic F127) was purchased from Sigma-Aldrich (Saint Louis, USA). Polyethylene glycol 300 (PEG300) and hydroxypropyl methylcellulose (HPMC, 15 Cps) were received from Sigma-Aldrich (Steinheim, German). Copovidone (Kollidon VA64) was purchased from BASF (Ludwigshafen, Germany). Distilled water was used in all experiments. All solvents used were of HPLC grade with the purity≥99.8%.
About PubCompare
Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.
We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.
However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
