Adiponectin and Estradiol Levels in Mice

After the 3T3-L1 cell culture step, adiponectin and estradiol levels in the medium and mouse plasma were determined using enzyme-linked immunosorbent assay (ELISA) kits (Mouse adiponectin/Acrp30 DuoSet® ELISA, DuoSet® ELISA Ancillary Kit 2, Estradiol Parameter Assay Kit, R&D systems, Inc., MN, USA). After the Ovariectomy and administration procedures steps, half of the mice in each group were euthanized by inhalation of isoflurane on day 20, and heart blood was immediately collected using a heparin-treated syringe. The blood was centrifuged at 4 °C and 4000 rpm for 15 min to obtain plasma, which was stored in a freezer at −80 °C. The plasma levels of adiponectin and estradiol were determined using an ELISA kit.
