Quantifying Bacterial GFP and Bioluminescence

For quantification of GFP and bioluminescence, overnight bacterial cultures of different strains with plasmid pGLhla were diluted in an appropriate medium containing 10 µg/mL chloramphenicol. Samples (100 µL) were transferred into a microtiter plate from culture tubes, and fluorescence was measured by using a DTX 800/880 multimode plate reader (Beckman). Bacteria with pGL plasmid were included as a control to allow correction for background fluorescence. For growth curve analysis, 100 µL of 106S. aureus strains were loaded to 96-well microtiter plates in triplicate and incubated at 37°C. The optical density at 620 nm (OD620), the fluorescence (GFP, Ex/Em: 485/535), and the bioluminescence were measured every 30 min in the DTX 800/880 multimode plate reader (Beckman).
