Nitric Oxide Quantification in Tissues

All procedures were performed following manufacturer’s protocol of nitric oxide assay kit (Abcam). The homogenizer (Tissue Miser Homogenizer; Fisher Scientific, Pittsburgh, PA, USA) was used to homogenize fresh tissues on ice. After collecting supernatant in a clean tube, perchloric acid (PCA, Fisher Scientific) was added in the tube for a final concentration of 1 M followed by incubation for 5 min. Then transfer the supernatant to a clean tube after centrifuge, adding an equal PCA volume of potassium hydroxide (KOH, Fisher Scientific) into the tube. Collecting supernatant after centrifuge for reaction wells preparation. A microplate colorimetric reader (OD540 nm, Bio-Rad, Hercules, CA, USA) was used to analyze absorbance. The NO volume was calculated dependent on a nitrite standard calibration curve.

Colorimetric Nitric Oxide Quantification

The nitric oxide assay kit (Abcam) was used to measure NO according to the manufacturer’s protocol. After standard curve preparation, fresh tissues were homogenized with a homogenizer (Tissue Miser Homogenizer; Fisher Scientific, Pittsburgh, PA) on ice. Supernatant was collected and transferred to a clean tube. A final concentration of 1 M perchloric acid (PCA, Fisher Scientific) was added to the supernatant tubes and then incubated for 5 min. The supernatant was transferred to a fresh tube after centrifuge. An equal PCA volume of potassium hydroxide (KOH, Fisher Scientific) was added to the supernatant. The supernatant was collected after centrifuge for 15 min. After reaction wells were assigned, a microplate colorimetric reader (OD540 nm, Bio-Rad, Hercules, CA) was used to measure the absorbance, converted into NO volumes on a nitrite standard calibration curve.
