Linsmaier and skoog ls medium

Manufactured by HiMedia

Linsmaier and Skoog (LS) medium is a widely used culture medium for the growth and maintenance of plant cells and tissues in vitro. It provides a balanced nutrient solution to support the nutritional requirements of plant cells, tissues, and organs. The medium contains essential macro and micronutrients, vitamins, and other growth-promoting substances necessary for plant cell culture.

Plants of Arabidopsis thaliana ecotype Columbia (Col-0) was used as a wild type (WT) and mutants and transgenics in this ecotype were generated and analyzed. Arabidopsis mutants used in this study have been previously described: cry1–30469 (link),cry2–18 , ubp12–1 (GABI_244E11), ubp12–2w (GABI_742C10), ubp13–1 (SALK_128312), ubp13–3 (SALK_130784), ubp12–2w ubp13–333 (link), and cop1–4.70 (link) Transgenic line cry2–1;UBQ10pro:9×Myc-6×His-3×Flag-CRY2 (FlashCRY2/CRY2oe) has been reported previously.4 (link) For all experiments, seeds were plated on 0.5× Linsmaier and Skoog (LS) medium (HiMedia Laboratories) with 0.8% agar, stratified for 2 days at 4°C in darkness and grown under the indicated light conditions in a LED growth chamber (Percival Scientific). Nicotiana benthamiana (tobacco) was grown under long days (16/8h day/night cycle) in the greenhouse or in a growth incubator at 22°C. Leaves of 3–4-week-old N. benthamiana were used for infiltration to transiently express proteins.

After surface sterilization, seeds were plated on 0.5× Linsmaier and Skoog (LS) medium (HiMedia Laboratories) containing 0.8% agar, stratified for 2 days in darkness at 4°C and then grown at 22°C under 100 μmol m−2 s−1 white light from a LED source in a growth chamber (Percival Scientific) unless otherwise specified.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
