THP-1 cells were seeded in a p96-well plate at a cellular density of 10,000 cells/well and treated with different concentrations of GSDMD45CRISPR-RhB-MSNs (0, 25, 50, and 100 µg/mL). Cells were incubated for 24 and 48 h, and then cell viability was determined by the WST-1 assay and inflammatory cell death by LDH activity assay (Promega, G7891, Madrid, Spain). For LDH activity, cell supernatants were analyzed by following the manufacturer’s instructions. As a positive control, 1% Triton-X100 was added.
Ldh activity assay
The LDH activity assay is a laboratory equipment product designed to measure the activity of the enzyme Lactate Dehydrogenase (LDH). LDH is an important enzyme involved in cellular metabolism, and its activity can be used as an indicator of cellular health and damage. The assay provides a quantitative measurement of LDH activity, which can be useful for various research and diagnostic applications.

About PubCompare
Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.
We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.
However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
