Characterization of A375 and U-2 OS Cell Lines

A375 melanoma (CRL-1619) and U-2 OS osteosarcoma (HTB-96) cells were from American Type Culture Collection. These cell lines were authenticated using STR analysis by Johns Hopkins Genetic Resources Core Facility and tested periodically for Mycoplasma using qPCR with negative results. The cells were maintained at 37°C in a humidified atmosphere containing 5% CO2. A375 cells were cultured in DMEM supplemented with 10% fetal bovine serum (FBS) and 4.5 g/L glucose and U-2 OS cells in DMEM with 15% FBS. The reagent used in this study was 12H-benzo[g]pyrido[2,1-b]quinazoline-4-carboxamide, N-[2(dimethylamino)ethyl]-12-oxo (BMH-21), which was synthesized as described by Colis et al. (2014b) (link) and verified for purity using liquid chromatography/mass spectrometry (LC/MS) and 1H nuclear magnetic resonance (NMR). MG132 was from Sigma-Aldrich and from Enzo LifeSciences.
