Mitotically inactivated buffalo FF cells (treated
with 10 μg/ml mitomycin-C) were cultured in T-25
flasks (Iwaki) with addition of ES medium for 7
days. Buffalo fibroblast-conditioned medium was
collected every day, centrifuged at 200 g for 3 minutes,
filtered with a 0.2 μm syringe filter (Millipore,
Watford, England) and frozen at -80˚C. After
thawing, the medium was equilibrated for 2 hours
at 5% CO2 and 37˚C and then used for the feederfree
culture of human ES cells.

Cell Culture Protocols for PAEC, PMVEC, PSM and PF

PAEC, PMVEC, PSM and PF cells were obtained from ScienCell (Germany) and grown in cell type-specific sterile growth medium (GM) in a humidified incubator at 37 °C and 5 % CO 2 in T25 flasks (Iwaki, Japan). In addition to supplied supplements, media were supplemented with: 200 μmol.l -1 glutamine dimer, 2 μg.ml -1 amphotericin-B, 60 μg.ml
penicillin-G and 100. μg ml -1 streptomycin sulphate (Invitrogen, UK). Medium was replaced aseptically every 3-4 days. Cells were sub-cultured every 10-14 days when ∼ 80-90 % confluent. Experiments were conducted at passage 4-8 at which point cells exhibited sufficient growth rates without loss of phenotype.
