Calu 3 cell line

Sourced in United States

The Calu-3 cell line is a human lung adenocarcinoma cell line derived from the pulmonary tissue of a 25-year-old Caucasian male. The cells exhibit an epithelial morphology and have been widely used in research to study lung physiology and disease processes.

Vero E6 cell line (Cat # CRL-1586, gender unknown) cell line was purchased from American Type Culture Collection (ATCC) and cultured at 37°C with 5% CO2in eagle’s minimal essential medium (MEM) supplemented with 10% fetal bovine serum (Invitrogen) and 1% penicillin/streptomycin and L-glutamine. Calu-3 cell line (Cat # HTB-55, obtained from a male, lung adenocarcinoma patient) was obtained from ATCC and maintained in EMEM+20%FBS at 37°C with 5% CO2.
The SARS-CoV-2 isolate hCoV-19/USA/MD-HP20874/2021 was obtained from BEI Resources, NIAID, NIH, and had been tittered on Vero E6 cells by plaque assay.

SARS-CoV-2 Isolate and Reporter Virus Propagation

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Vero E6 cell line (ATCC CRL-1586) and the Caco-2 cell line (ATCC HTB-37) were purchased from the American Type Culture Collection (ATCC) and cultured in Eagle’s minimal essential medium (EMEM) supplemented with 10% fetal bovine serum (FBS; Invitrogen), 1% penicillin-streptomycin, and l-glutamine. The Calu-3 cell line (ATCC HTB-55) was obtained from the ATCC and maintained in EMEM plus 20% FBS. The Huh7.5.1 cell line was a gift of F. Chisari and was maintained in Dulbecco’s modified Eagle’s medium (DMEM) plus 10% FBS (41 (link)).
The SARS-CoV-2 isolate USA-WA1/2020 was obtained from BEI Resources, NIAID, NIH, and had been passed three times on Vero cells and one time on Vero E6 cells prior to acquisition. It was further passaged once on Vero E6 cells in our laboratory. The virus has been sequence verified to contain no mutation in its original seed virus. A reporter virus (icSARS-CoV-2-mNG) that contains an insert of monomeric neon green (mNG) in the SARS-CoV-2 USA-WA1/2020 backbone was obtained from Pei-Yong Shi’s laboratory (University of Texas Medical Branch) at passage 2 and was further expanded once on Vero E6 cells (28 (link)) at the FDA.

Vero E6-TMPRSS2-cells were maintained in DMEM Medium (gibco) containing 10 % FBS and 1 mg/ml G418 Sulfate and 1% Penicillin-Streptomycin. The Calu-3 cell line was purchased from ATCC (HTB-55), and maintained in Dulbecco’s Modified Eagle Medium/Nutrient Mixture F-12 (DMEM/F-12) supplemented with 10% of Fetal Bovine Serum (FBS) and 1% of 5000 units/ml Penicillin and 5000 μg/ml. The WT SARS-CoV-2 (HKU-001a strain, GenBank accession number MT230904) and SARS-CoV-2 B.1.1.529/Omicron (GISAID accession number EPI_ISL_7138045) strains were isolated from respiratory tract specimens of laboratory-confirmed COVID-19 patients in Hong Kong and cultured in Vero E6-TMPRSS2 cell32 (link).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
