purchased from SHOWA, Japan. Disodium phosphate (Na2HPO4, 99%), monosodium phosphate (NaH2PO4, 99%), hydrogen peroxide (H2O2, pure, 30%
w/w), and Tris base were purchased from Sigma-Aldrich. Anti-IL-6 antibody

purchased from Fisher. Ethanol (99.5%) was purchased from ECHO. Tween
20 was bought from Merck, Germany. Human oral cancer tissue array
(product number T271b), oral cancer tissue array (product number T273),
and human oral cancer tissue slices (product numbers HuCAT481 and
HuCAT503) were purchased from Biomax. Details of human oral cancer
samples are presented in the Supporting Information (SI)
and sex of the pathology sites. Deionized water was obtained from
Sartorius (18.2 MΩ·cm). The conductive silver paint was
bought from Pelco. Polyimide was purchased from Kapton (DuPont). Parafilm
was purchased from Parafilm M (Merck, Germany). Gold wire (25 μm)
was purchased from Tanaka Electronics, Singapore. Sylgard 184 A&B
was bought from Uni-Onward, Taiwan.
Insulin Enzyme-Linked Immunosorbent Assay
medium (DMEM), fetal bovine serum (FBS), Penstrep, and trypsin-EDTA
were obtained from Gibco. Monosodium phosphate (NaH2PO4), Rat/Mouse
(EZRMI-13K) Insulin ELISA Assay Kit, and ethanol were purchased from
Merck. Magnesium chloride (MgCl2), glucose, calcium chloride
(CaCl2), potassium chloride (KCl), HEPES, monopotassium
phosphate (KH2PO4), sodium chloride (NaCl),
dipotassium phosphate (K2HPO4), sodium bicarbonate
(NaHCO3), formaldehyde, 2-propanol, BSA, and Bradford were
purchased from Himedia, India. Glucose oxidase, 4-aminoantipyrine N-ethyl-N-sulfopropyl-M-toluidine, horseradish peroxidase, disodium phosphate (Na2HPO4), and chloroform were purchased from Sigma, India.
Trizol was obtained from Ambion Life Technology.

standard solution (0.1 M, NaOH), poly(acrylic acid) (PAA, Mw ∼ 1.2 kDa), branched polyethylenimine
(BPEI, Mw = 750 kDa, Mn = 60 kDa), poly(methacrylic acid) (PMAA, Mw = 60 kDa), poly(4-vinylpyridine) (P4VP, Mw = 160 kDa) glutaraldehyde, sodium acetate buffer (NaOAc),
sodium dihydrogen phosphate (NaH2PO4), and disodium
phosphate (Na2HPO4) were obtained from Sigma-Aldrich.
Hydrochloric acid (1 N standard solution, HCl) was purchased from
Acros Organics. Hydrogen peroxide (30% in water, H2O2) was obtained from Fisher Scientific Inc. Cross-linkable
quaternized poly(4-vinylpyridine) (qP4VP, 160 kDa) was synthesized,
as described in section 2.3. All chemicals were used without further
purification steps. All solutions were prepared using ultrapure water
(Milli-Q, <18.2 MΩ cm). For LbL deposition, qP4VP and PAA
solutions were prepared at a concentration of 0.2 mg/mL in 10 mM sodium
phosphate buffer and adjusted to pH 6.5. The salinity solutions were
prepared by adding Sodium chloride powder to the phosphate buffer
solutions at pH 8.1.

Analytical LC-MS Reagent Preparation
Formic acid (FA) (LC-MS grade) and ammonium acetate (AA) (analytical reagent grade) were obtained from Fisher scientific (Waltham, MA, USA). Sodium bicarbonate was from Laurylab (Brindas, France) and hydrochloric acid from Carlo Erba Reagents (Val-de-Reuil, France). Water was purified by an Elga purification system (Veolia, Paris, France). Mobile phases (all proportion expressed as v/v) were filtered through 0.2 µm PTFE membranes (Merck, Germany) before use.
