Lymphatic Organ Characterization in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
At week 40, all groups of mice from both studies were euthanized to collect lymphatic organs such as the spleen and lymph nodes. Then, the cell suspensions of these secondary lymphoid organs were labeled with anti-mouse CD4 PE, anti-mouse CD8a FITC, anti-mouse CD45R (B220) FITC, anti-mouse CD62L APC and anti-mouse CD27 APC (eBioscience). The procedure was conducted according to the manufacturer’s protocol [31 ]. The labeled cells were then analyzed by flow cytometric analysis using a BD Accuri® C6 flow cytometer.

Hematopoietic Stem Cell Mobilization and Neutrophil Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole blood samples were red cell lysed and analyzed using the BD LSRII system with FACS software (BD, Franklin Lakes, NJ).
For enumeration of HSC mobilization (Lin-Sca-1+c-kit+Flt3 or LSKF cells) [7 (link)], cells were stained with Mouse Lineage Panel (BD, Franklin Lakes, NJ), streptavidin PE (eBioscience, San Diego, CA), anti-mouse Sca-1 PE-Cy7 (eBioscience, San Diego, CA), anti-mouse c-kit FITC (eBioscience, San Diego, CA), anti-Flt3 APC (eBioscience, San Diego, CA), and 7-ADD (BD, Franklin Lakes, NJ).
For evaluation of neutrophil L-selectin shedding, cells were stained with anti-mouse Gr-1 FITC (eBioscience, San Diego, CA), anti-mouse CD115 PE (eBioscience, San Diego, CA), anti-mouse CD62L APC (eBioscience, San Diego, CA), and 7-AAD (BD, Franklin Lakes, NJ).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
