Organic Black Quinoa Phytochemical Analysis

Dehulled seeds of organic black quinoa (Chenopodium quinoa Willd.), labelled as originating from Bolivia (South America) and packed in a protective atmosphere, were purchased in a retailer specialized in organic and biodynamic products in Italy. Proximate composition per 100 g was, as reported in the nutrition label: 380 kcal energy; 6.2 g fat, of which 0.7 g saturates; 65 g carbohydrates, of which 4.0 g sugars; 9.0 g fibre; 12 g protein; and 0.01 g salt.
Folin–Ciocalteu’s Reagent, methanol, ethanol, acetone, acetic acid and acetonitrile were purchased from Carlo Erba Reagents (Milan, Italy).
Caffeic acid, (+)-catechin, o-coumaric acid, p-coumaric acid, (−)-epicatechin, 3,4-dihydroxycinnamic acid, t-ferulic acid, gallic acid, gentisic acid, 4-hydroxybenzoic acid, protocatechuic acid, rutin, sinapic acid, syringic acid, vanillic acid, and vanillin were purchased from Extrasynthèse (Geney, France) and Sigma-Aldrich (St. Louis, MO, USA).
HPLC grade solvents and water purified by a Milli-Q system (Millipore Corp., Billerica, MA, USA) were used in HPLC analysis.

Standardized Phytochemical Characterization

The standards of 4-hydroxybenzoic acid, p-coumaric acid, ferulic, isoferulic acid, pinobanksin, chrysin, sakuranetin, naringenin, apigenin, kaempferol, isorhamnetin, acacetin and pinocembrin were obtained from Extrasynthese (Genay, France) and galangin from Alfa Aesar, (Haverhill, MA, USA). The antibiotics: ampicillin, oxacillin, teicoplanin, gentamicin, amikacin, fusidic acid, erythromycin, mupirocin, rifampicin, levofloxacin, norfloxacin, linezolid, bacitracin were purchased from Argenda (Poland). Methanol, ethanol, vanillin, benzoic acid, cinnamic acid were purchased from (POCH, Gliwice, Poland). Ultrapure H2O (18.0 MΩ) was obtained with a Milli-Q Advantage A10 system (Millipore, Billerica, MA, USA). The absorbance of the reaction mixture in Folin-Ciocalteu, DPPH assays were measured using a Genesys 20 spectrophotometer (Thermo Scientific, Waltham, MA, USA).

Standards of 33-α-L-plus 23-α-L-arabinofuranosyl-xylotetraose (XA3XX/XA2XX), 23,33-di-α-L-arabinofuranosyl-xylotriose (A2, 3XX), and xylotriose were obtained from Megazyme (Wicklow, Ireland). Raffinose and cellobiose standards were supplied by Merck (Darmstadt, Germany).

Standards of compounds (ferulic acid, caffeic acid, phenylalanine, catechin, apigenin, quercetin, isoorientin, apigenin 6-C-glucoside-8-C-arabinoside, apigenin 6,8-di-C-glucoside, 4-hydroxybenzoic acid, luteolin-3,7-di-O-glucoside, tricin 7-O-glucoside, abscisic acid) were purchased from Extrasynthese (Genay, France). isoorientin 2”-O-glucoside, isovitexin 7-O-glucoside, isoscoparin 2”-O-glucoside, and apigenin 6-C-arabinoside-8-C-glucoside were purified from plant material and their structures were confirmed with NMR analysis as described previously [13 (link)]. Standards of zearalenone, tryptophan, 2,2-diphenyl-1-picrylhydrazyl, and ascorbic acid were obtained from Merck SA. Hypochloride was obtained from BioShop (Burlington, ON, Canada).

HPLC Analysis of Phenolic Compounds

Methanol and acetonitrile (both HPLC grade) were acquired from Fisher Chemical (Leicestershire, UK). Formic acid (98+%, pure, analytical reagent grade) was obtained from Acros Organics (Geel, Belgium).
Ultrapure water with a resistivity of 18.2 MΩ•cm at 25 °C (Milli-Q, Waters) was used as solvent and for mobile phase preparation.
Trans-cinnamic acid (CA) (99.8%), and caffeic acid (99.3%) were obtained from Sigma-Aldrich (St. Louis, MO), while 4-hydroxybenzoic acid (100%), aucubin (98.6%) and verbascoside (95.1%) were from Extrasynthese (Genay, France). Methylparaben and propylparaben were purchased from Adipogen (Liestal, Switzerland). Conical centrifugal filters (modified PES 3K, 500 μL) were obtained from VWR (Radnor, USA).
