Pharmacokinetic Study of CTE, NGTS, and CNP

CTE, NGTS, and CNP powders were dissolved using saline, and were orally dosed at 50 mg·kg−1, 60 mg·kg−1, and 50 mg·kg−1 CTE + 60 mg·kg−1 NGTS, respectively, whereas saline was administered to the vehicle group. Six rats were used in each group, and all rats fasted overnight but had free access to water prior to treatment. A terminal sampling design was used to collect blood samples at 0 (pre-dose), 0.08, 0.16, 0.25, 0.5, 1, 2, 3, 4, 6, 8, 12, 24, 48, 72, and 96 h. At each time, 0.5 mL of blood was collected in the heparin sodium tubes. Plasma was separated by centrifugation at 4000 rpm for 10 min and stored below −80 °C until bioanalysis. Oasis PRiME HLB SPE cartridges (1 cc/30 mg, Waters, Milford, MA, USA) were used to process the plasma samples (Supplementary Materials).

Synthesis and Purification of Phenanthrenequinones

1,2-Phenanthrenequinone (1,2-PheQ) and 3,4-phenanthrenequinone (3,4-PheQ) were prepared as previously described (Figure 3) (Magdziak et al., 2002 (link)).) (Indianapolis, IN, USA). Oasis® PRiME HLB SPE cartridges (200 mg/6 mL/# 186008057) were from Waters (Milford, MA, USA). A Bond Elut C18 96 square-well plate (100 mg/2 mL/# A396011C) was purchased from Agilent (Santa Clara, CA, USA). All solvents used in this study including hexane, isopropanol, acetonitrile, ethyl acetate and methanol were HPLC grade and obtained from Fisher Scientific (Fair Lawn, NJ, USA). Ultrapure H2O from a Millipore Milli-Q Advantage A10 Water Purification System was used throughout the study.

Chromatographic Separation and Immunoassay of Mycotoxins in Feed

The reagents used in the chromatographic separation of mycotoxins and their extraction from feed samples were: acetonitrile (ACN) and methanol (MeOH), both of gradient grade and purchased from Honeywell Riedel-de Haën (Muskegon, MI, USA); chloroform and water, also liquid chromatography (LC) gradient grade, provided by Fisher Scientific (Bishop Meadow, UK); and pro-analysis grade orthophosphoric acid obtained from Panreac (Barcelona, Spain). In addition, OASIS PRIME HLB SPE cartridges were obtained from Waters (Milford, MA, USA). In the case of DON and STER analysis, commercial enzyme-linked immunosorbent assay (ELISA) kits suitable for mycotoxin quantification in feed were used. For DON analysis, the RIDASCREEN® FAST DON SC kit (Art. No. R5905 R-Biopharm AG, Darmstadt, Germany) was employed; while for STER quantification, the Creative-diagnostic Sterigmatocystin DEIANJ08 ELISA kit was selected (NY, USA). In both cases, all the reagents needed were included in the kits.
