Evaluating Aortic Endothelial Cell Response

C3H/HeJ (C3H) and C57BL/6J (BL6) mice were purchased from the Jackson Laboratory. Abcc6-Tg mice on C3H background were generated as described [8] (link) and contain an Abcc6 BAC transgene derived from C57BL/6J. Abcc6 knockout (Abcc6−/−) mice were obtained from Dr. A.A.B. Bergen [4] (link) and backcrossed for 10 generations on a C57BL/6J background. Littermates were used as wild type controls. All mice were fed a standard chow diet (Diet 8604, HarlanTeklad, Laboratory). All mice were used for experiments at 3–4 months of age.
Bovine aortic endothelial cells (BAEC) were cultured as previously described [16] (link), and BMP4 (0–40 ng/ml, R&D Systems) in culture medium with 10% fetal bovine serum (FBS), or serum from C3H or C3H-Abcc6Tg mice was added at the time of plating.

Constant Nutrition Mouse Acclimatization

Ninety-six, 5–6 week old male C3H/HeJ (C3H) and C57BL/6J (B6) mice (The Jackson Laboratory, Bar Harbor, ME, USA) were placed on a constant nutrition no fluoride diet #5861 [Test Diet®, Richmond, IN, USA; fluoride: 0ppm, calcium: 0.60%, phosphorus: 0.57%, Vit D3: 2.2 IU/g, and Energy: 4.09 (kcal/g)2] and provided distilled drinking water, ad libitum, for one week to acclimatize. Mice were housed in The Division of Lab Animal Medicine facility at The University of North Carolina at Chapel Hill, an AAALAC accredited unit. Mice were placed in 12:12 light and dark cycles at 21°C ambient temperature. All procedures involving vertebrate animals were approved by the Institutional Animal Care and Use Committee at The University of North Carolina at Chapel Hill.

Mouse Strain Comparison for SLE Research

Comparative Analysis of BMAT in C3H and B6 Mice

The Institutional Animal Care and Use Committee (IACUC) at Washington University in St. Louis approved all procedures, and these experiments were performed in AAALAC accredited facilities. For all experiments, C3H/HeJ (C3H, Stock:000659) and C57BL/6J (B6, Stock:000664) mice were obtained from Jackson Labs; mice were acclimatized for 1 week prior to experiments. Mice were housed on a 12-h light/dark cycle at 70 ± 2 degrees Fahrenheit and fed standard chow (LabDiet® 5053). Relative to B6 mice, C3H mice are known to have a significant expansion of BMAT in the proximal tibia by 12-weeks of age (29 (link)).

Genetic knock out mice on the BL6 background purchased from Jackson include: B6.129P2-Tcrbtm1Mom/J (TCRβKO), B6.129P2-Tcrdtm1Mom/J (TCRδKO), and B6.129S7-Rag1tm1Mom/J (RAG1KO). Mice were bred in our animal facility, given ad libitum water and food, and housed under specific pathogen-free conditions. All mice were at least 8 weeks old and age- and sex-matched for each experiment.

Strain-Specific Effects of Nicotine on Learning

Male A/J (A; n = 19), BALB/cByJ (BALB; n = 18), C3H/HeJ (C3H; n = 18), C57BL/6J (B6; n = 18), CBA/J (CBA; n = 18), DBA/1J (D1; n = 20), and DBA/2J (D2; n = 18) mice were obtained from Jackson Laboratory (Bar Harbor, ME). 129/SvEv (129) mice were obtained from Taconic Farms (Germantown, NY). These inbred strains were chosen because previous work from our lab found phenotype differences in these strains for the effects of nicotine on learning and memory (Portugal, Wilkinson, Kenney, Sullivan, & Gould, 2012 (link)). Mice were trained at 8–10 weeks of age. Mice were housed in groups of three per cage and maintained on a 12-hr light–dark cycle (lights on 7:00 a.m.) with unrestricted access to food and water. Experiments took place during the light phase. All behavioral and surgical procedures were approved by the Temple University Institutional Animal Care and Use Committee.

Breeding Stocks of Common Mouse Strains

Breeding stocks of wildtype male and female ICR (CD-1) mice (Harlan Laboratories, Livermore, CA), C57BL/6J (B6), and C3H/HeJ (C3H) (Jackson Laboratory, Bar Harbor, ME) were maintained in our laboratory in a pathogen-free facility. C3H/B6 F1 hybrids were generated by mating several C3H/HeJ females to a single C57BL/6J male. W/Wv males used as germ cell transplantation recipients were generated from breeding heterozygous W and Wv C57BL/6J mice (Jackson Laboratory, Bar Harbor, ME). Animals were housed in polysulfone cages (Allentown Inc. Allentown, NJ, Jag 75 micro isolator model) on ventilated racks, in cages containing Sanichip 7090A bedding (Harlan Laboratories) and enrichment material—a nestlet (Ancare, Bellmore, NY), and a Diamond Twist enrichment product (Harlan Laboratories). Drinking water and chow (Purina Lab Diet, 5K52) were provided ad libitum. Individually-housed male animals received a single pellet of Teklad Global 19% Protein Extruded Rodent Diet once a week as an enrichment product (Harlan Laboratories).
