Immunohistochemical Scoring of GPSM2 and DYNC1I1

Check if the same lab product or an alternative is used in the 5 most similar protocols
IHC staining was performed as described previously (19 (link)). Anti-human GPSM2 rabbit antibody was used at a dilution of 1:100 (ab84571; Abcam, Cambridge, UK); Anti-human DYNC1I1 mouse antibody was used at a dilution of 1:100 (ab23905; Abcam, Cambridge, UK); phosphate-buffered saline was used as a negative control. Each section was evaluated and scored independently by two pathologists. A semi-quantitative scoring system was used in this assay. Intensity was scored as “0” (negative), “1” (weak), “2” (moderate), and “3” (strong), and the percentages of tumor cells within each category were calculated. The percentage score was multiplied by the staining intensity score to generate the IHC score. The histological score range was from zero (minimum) to 300 (maximum). Positive expression was defined as detectable immunoreactions with an IHC score > 10.

Immunohistochemical Evaluation of GPSM2

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immunohistochemical staining was carried out as described previously.17 Staining was carried out with an anti‐human GPSM2 rabbit Ab that was used at a dilution of 1:100 (ab84571; Abcam); PBS was used as a negative control. Each section was evaluated and scored independently by 2 pathologists. A semiquantitative scoring system was used in this assay. Intensity was scored as: 0, negative; 1, weak; 2, moderate; and 3, strong. We also calculated the proportion of tumor cells within each category. The proportional score was then multiplied by the staining intensity score to generate a final immunohistochemistry (IHC) score. The IHC scores ranged from 0 (minimum) to 300 (maximum). Positive expression of GPSM2 was defined as detectable immunoreactions with an IHC score greater than 10.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
