Quantification of Phenolic Compounds

All solvents were HPLC grade and were obtained from Sigma–Aldrich or Fisher Scientific. While we acknowledge that avenanthramides are phenolic alkaloids, the terms phenolics is used throughout the paper to include both avenanthramides and phenolic acids.

Detailed Polyphenolic Compound Preparation

Gibberellic acid, catechin, syringaldehyde, sinapinic acid, isovanillic acid, gentisic acid, ferulic acid, luteolin, quercetin, kaempferol, naringenin, chlorogenic acid, p-coumaric acid, vanillic acid, rosmarinic acid, caffeic acid, protocatechuic acid and gallic acid were obtained from Sigma (St. Louis MO, USA). The chemical structures of eighteen polyphenolics studied are listed in Table 1.
Methanol (MeOH, HPLC grade) was purchased from Fisher Scientific (Pittsburgh, PA, USA). Sodium tetraborate (Na 2 B 4 O 7 ), boric acid (H 3 BO 3 ) sodium hydroxide (NaOH), butylated hydroxyanisole (BHA) and hydrocloric acid (HCl) were obtained from Sigma (St. Louis MO, USA).
Stock solutions of polyphenols were prepared by dissolving enough of the studied compound in MeOH (50 mL) to give a 100 ppm in solution. Borate buffer (40 mM) was prepared by mixing Na 2 B 4 O 7 with H 3 BO 3 (500 mM) in ultrapure water. All used reagents were protected from light. The prepared solutions were stored at 4 °C for several days and at -18 °C for long-term storage.

HPLC/MS Analysis of Lignin Markers

The ultrapure water (18.2 M cm, 0.01 TOC) was produced by a Purelab Flex (Elga, High Wycombe, U.K.) and formic acid (98%) was obtained by Fluka (Sigma Aldrich, Buchs, Switzerland). Levoglucosan (purity 99%), vanillin (VAN) (≥ 98%), syringic acid (SyA) (≥ 95%), homovanillic acid (HA) (≥ 98%), isovanillic acid (IVA) (97%), p-coumaric acid (PA) (≥ 98%), coniferyl aldehyde (CAH) (98%), were purchased from Sigma Aldrich, vanillic acid (VA) (≥ 97%), syringaldehyde (SyAH) (≥ 97%), ferulic acid (FA) (≥99%) from Fluka. Levoglucosan 13 C6(98% isotopic enriched, ≥ 98% chemical purity) from Cambridge Isotope Laboratories Inc. (Andover, MA), vanillic acid 13 C1(98% isotopic enriched, ≥ 98% chemical purity) and vanillin 13 C6(98% isotopic enriched, ≥ 98% chemical purity) from Sigma Aldrich.
