Paeoniflorin Effects on RA-FLS Cells

Human fibroblast-like synoviocyte (H-FLS) and RA-FLS cell lines (Jennio Biotechnology, Guangzhou, China) were incubated with Dulbecco's modified Eagle's medium (DMEM; Baikaimei Biotechnology, Shenzhen, China) including 10 % fetal bovine serum (FBS; Procell Life Science&Technology, Wuhan, China) and 1 % penicillin/streptomycin (P/S; Baikaimei Biotechnology, Shenzhen, China). Cells were routinely grown in the humidified condition at 37 °C and 5 % CO2. For paeoniflorin treatment, RA-FLSs were exposed to various doses (0, 25, 50, 75, 100 and 200 μg/ml) of paeoniflorin (Yuanye Bio-Technology, Shanghai, China) for 24 and 48 h.

BZXD Marker Component Analysis

According to the Pharmacopoeia of the People's Republic of China (2020 edition), six main components in BZXD have been identified. The quality stability and repeatability of these six marker components were detected by UPLC-MS/MS analysis method. We purchased the standard reference materials of Rosmarinic acid, Paeoniflorin, Glycyrrhizic acid, Salvianolic acid B, p-Coumaric acid, and Ferulic acid from Yuanye Bio-Technology Co., Ltd. (Shanghai, China). Qualitative analysis was carried out using an AB Sciex API 4000 LC/MS/MS system. Analytes were separated using a linear gradient program at a flow rate of 0.30 ml/min using a mobile phase consisting of 2 mmol/l ammonium acetate in water (A) and acetonitrile (B).
Precisely pipette 0.200 ml of thawed BZXD concentrate at 4 °C into a 2 ml brown PE tube, 0.6 ml mass spectrometry grade acetonitrile solution, and vortex for 5 min. Centrifuge at 13000r/min at 4 °C for 20 min. Take the supernatant and dilute it by 4 times, shake and mix, and filter through a 0.22-micron filter to be tested. The samples were detected and analyzed by negative ion electrospray tandem mass spectrometry (Supplementary Figure S1).

The primary antibody of rabbit anti-CXCL2 (bs-1162R) was purchased from Bioss Inc., (Beijing, China) and GAPDH, 1:4,000 (14C10) was from Cell Signaling Technology (Beverly, MA, United States). The secondary antibody Goat anti-Rabbit IgG (1:4,000) (ZB-2301) was purchased from ZS bio (Beijing, China). 2, 2, 2-Tribromoalcohol (Cat#:T48402) and Bacterial lipopolysaccharide (Cat#: L2880-10 MG) were purchased from Sigma Aldrich (St. Louis, MO, United States). Rhod2-AM (Cat#: zy0129) and Anti-GRP78 (BiP) antibody (ab21685) were purchased from Abcam (Cambridge, MA, United States).

Quantitative Analysis of Bioactive Compounds

Quantitative Analysis of Shanzhiside and Geniposide in GF Extract

A reference standard of shanzhiside (purity 98%) was purchased from Shanghai Tauto Bio-tech Co., Ltd. (Shanghai, China). A reference standard of geniposide (purity 98%) was purchased from Shanghai ANPEL Laboratory Technologies Inc. (Shanghai, China). Reference standards of genipin-1-gentiobioside and genipin (purity 98%) were purchased from Dalian Meilun Biotechnology Co., Ltd. (Dalian, Liaoning, China). Paeoniflorin (purity 98%) was purchased from Shanghai Yuanye Bio-technology Co., Ltd. (Shanghai, China) and used as an internal standard (IS). GF extract was prepared in our lab and the chemical analysis was performed as our previous research (The proportions of shanzhiside, genipin-1-gentiobioside and geniposide were 0.06%, 3.75% and 12.45%, respectively) [27 ]. The herbal materials of SSP were purchased from Chengdu and authenticated by Dr. Luping Qin from the Second Military Medical University. Acetonitrile, methanol (analytical grade) and acetic acid (HPLC grade) were purchased from Merck Company (Darmstadt, Germany) and Tedia (Fairfield, USA), respectively. Deionized water (18.2 MΩ/cm) was generated in-house using a Milli-Q System from Millipore (Bedford, MA, USA).

Inflammatory Response Modulation Protocol

Rat peripheral blood mononuclear cell (PBMC) isolation kit, TaqMan RT-qPCR kit, and cDNA synthesis kit were purchased from Solarbio Technologies (Beijing, China). All the solvents were of analytical grade and supplied by Merck Chemicals (Shanghai, China). High-purity compounds (>98%) kaempferol, paeoniflorin, matrine, sophocarpine, and sinomenine were all obtained from Yuanye Bio-Technology (Shanghai, China).

Authentication of Herbal Medicines in MZRW

All the six herbal medicines (HMR: RM130121-08; DH: 130524; BS: RM130128-11; KXR: RM121026-04; HP: RM121026-05; ZS: RM121210-11) contained in MZRW were supported and authenticated by PuraPharm Company, Nanning department (Nanning, China). The reference standards of gallic acid, amygdalin, paeoniflorin, hesperidin, neohesperidin, naringin, magnolol, honokiol, aloe emodin, physcion, chrysophanol were purchased from Shanghai Yuan-Ye Bio-technology company (Shanghai, China). Emodin, rhein, albiflorin were purchased from the National Institute for the Control of Pharmaceutical and Biological Products (Beijing, China). The purity of all these reference standards was ≥98.0% (HPLC). HPLC grade of acetonitrile, methanol and formic acid were purchased from Merck (Darmstadt, Germany). Deionized water was purified by the Millipore water purification system (Millipore, Milford, MA, United States). All other reagents used were of analytical grade. Twelve male Sprague-Dawley rats (220–250 g) were kindly provided by the Experimental Animal Center of Chinese University of Hong Kong (Hong Kong). The rats were bred in an environmentally controlled room (22 ± 2°C, relative humidity 50 ± 20%) with a natural light-dark cycle for 7 days before the experiment carried out. The animal study was carried out in accordance with the Guideline for Animal Experimentation of Hong Kong.

HPLC-grade acetonitrile and methanol were procured from Tedia (Fairfield, OH, United States), and formic acid (LC-MS grade) was purchased from Aladdin (California, United States). All other reagents were of analytical grade.
