Cy5.5-labeled Peptide Synthesis

The peptides were synthesized using a PS3 automatic synthesizer (Protein Technologies Inc, Tucson, AZ) and labeled with Cy5.5, a near-infrared red (NIR) fluorophore (cyanine5.5 NHS ester, #27020; Lumiprobe, Hunt Valley, MD ), on the C-terminus using a GGGSK linker (14 (link)). The peptides were purified to >99% by HPLC and lyophilized for storage at −80 °C.

Purification and Analysis of Recombinant Proteins

Visualizing aPDL1 Accumulation in Metastatic Lungs

To study aPDL1 accumulation in metastasis-bearing lung tissues, aPDL1 was labelled by Cyanine5.5 NHS-ester (Lumiprobe) before conjugated to the platelets. After purification, the Cy5.5-labelled aPDL1, P@aPDL1, and Vadimezan + P@aPDL1 were administrated to the mice intravenously (10 days after 4T1 cells inoculation). Twenty-four hours post-administration, lungs were collected, then the aPDL1 in the lungs were detected by IVIS, the fluorescence signals were analysed by Living Image Software.

MIT was supplied by Selleckchem. Cyanine5.5 NHS ester was purchased from Lumiprobe Corporation and luciferin was from Caliper LifeScience.

All fluorescent dyes (FDs) are presented in Table S1.
Water was purified by a Simplicity 185 water system Millipore (Burlington, MA, USA) and had a resistivity of 18.2 MΩ·cm at 25 °C. The deionized water was used to prepare buffers and oligonucleotide solutions.

LIVE/DEAD® fixable green dead cell stain was purchased from ThermoFisher. AlexaFluor® 680-Wheat Germ Agglutinin conjugate (WGA) and LysoTracker® Blue DND-22 were purchased from Life Technologies. Cell culture reagents such as DMEM, FBS, trypsin, and PBS were purchased from Atlanta Biologicals. Ultrapure water (18 MΩ) was used for preparation of all buffers and in all experiments. All solvents were of analytical grade, purchased from commercial sources and used without further purification, except DMF which was dried over CaH2 under N2, filtered, and distilled under reduced pressure.
