Hypoxia Response Element Assay

The eLUCidate HRE reporter cell line, a stably transfected HeLa cell line which expresses Renilla luciferase reporter gene under the transcriptional control of the hypoxia response element (HRE), was obtained from Genlantis (San Diego, CA). The cells were seeded in a 96-well plate at 1 × 104 cells per well and treated with various exosomes at 50 μg/ml for 48 h. The HRE signals were detected by using a Renilla Luciferase Glow Assay Kit (Thermo Fisher Scientific). Briefly, the cells were rinsed with PBS once. Cell lysis buffer was added at 50 μl/well followed by shaking on a shaker for 15 min.
