Hepatocyte Organoid Toxicity Assay

PHH-derived organoids were seeded on a 10 mg/mL HYDROX-coated 96-well plate. After 14 days of the HYDROX culture, Org-HYDROX were cultured with organoid expansion medium containing different concentrations of acetaminophen (Wako), troglitazone (Wako) and amiodaron (FUJIFILM Wako Pure Chemical) for 7 days. As a control group, PHHs (lot OHO; Celsis) cultured with hepatocyte culture medium (HCM; Lonza) for 48 h after plating on a 96-well plate were exposed to different concentrations of acetaminophen (Wako), troglitazone (Wako) and amiodaron (FUJIFILM Wako Pure Chemical) for 7 days. Cell viability was evaluated using a Cell Counting Kit-8 (Dojindo Laboratories) based on a WST-8 assay according to the manufacturer's instructions. The absorbance at 450 nm was determined by a multiplate reader (Bio-Rad). Cell viability was calculated as a percentage of the control (DMSO-treated group) viability, which was taken as 100%.

Cytotoxicity Evaluation of Drugs

Acetaminophen and Rotenone Effects in Sprague-Dawley Rats

The following experiments were approved by the Ethical Committee of the Central Research Laboratory, Hamamatsu Photonics. Control male Sprague-Dawley rats (8 weeks of age; 260–280-g body weight) were purchased from Japan SLC, Inc. (Hamamatsu, Japan). Acetaminophen was from Wako Pure Chemical Industry (Osaka, Japan). Isoflurane and rotenone were purchased from Dainippon Pharmaceutical (Osaka, Japan) and MP Biochemicals LLC (Illkirch, France), respectively. The precursor of 18F-BCPP-BF and its standard compound were obtained from the NARD Institute (Amagasaki, Japan).

Synthesis and Characterization of Coumarin Derivatives

Synthesized oligonucleotides were obtained from Integrated DNA Technologies (Coralville, IA).
Coumarin, phenacetin, 7-hydroxyCoumarin, and acetaminophen were obtained from Fujifilm Wako Pure Chemicals (Osaka, Japan). Pooled liver microsomes of humans (75 males and 75 females), 3 male and 3 female dogs, and 3 male and 3 female minipigs were purchased from Corning Life Sciences (Woburn, MA). Liver microsomes were prepared from two tree shrews as described previously (Yamazaki et al., 2014; Uno et al., 2022a; Uno et al., 2022c) . Recombinant human CYP2A6 and CYP2A13 in bacterial membranes were prepared using the bicistronic protein expression system with human NADPH-P450 reductase as described previously (Shimada et al., 1997; Uehara et al., 2015; Uno et al., 2006; Uno et al., 2022a; Uno et al., 2022c) . All other reagents were purchased from Sigma-Aldrich or Fujifilm Wako Pure Chemicals, unless otherwise specified.

Louis, MO). Acetaminophen, acetic acid, sodium phosphate, and ammonium acetate solution were purchased from Wako Pure Chemical Industries Ltd. (Osaka, Japan). β-estradiol-3-β-Dglucuronide sodium salt was obtained from Santa-Cruz Biotechnology Inc. (Santa-Cruz, CA).
All chemicals used for high-performance liquid chromatography (HPLC) and mass spectrometry (MS) were HPLC or MS grade and were obtained from Kanto Chemical Co. Inc.
(Tokyo, Japan).
