Characterization of MT-102 Lipid Formulations

MT-102 was obtained from MTHERA PHARMA (Seoul, Republic of Korea). Olive oil, corn oil, and soybean oil were purchased from Daejung Chemicals and Materials Co., Ltd. (Gyeonggi-Do, Republic of Korea). Transcutol HP, Masine CC, Peceol, Capryol 90, Labrafil M 1944 CS, Labrafil M 2125CS, and Labrasol were purchased from Gattefossé (Saint-Priest, Lyon, France). Kolliphor RH 40, Kolliphor HS 15, and Kolliphor ELP were obtained from BASF (Ludwigshafen, Germany). Capmul PG8, Captex 355, and Capmul MCM C8 were provided by ABITEC Corporation (Janesville, WI, USA). Tween 80, ethanol, polyethylene glycol 200 (PEG 200), propylene glycol, and 6-methoxyflavone were purchased from Sigma Aldrich (St. Louis, MO, USA). Dextran sulfate sodium salt (colitis grade) was purchased from MP Biomedicals (Solon, OH, USA). All solvents used for HPLC analysis were purchased from Merck KGaA (Darmstadt, Germany).

Formulation and Characterization of MT-102

MT-102 was provided by MTHERA PHARMA (Seoul, Korea). Indirubin, 6-methoxy flavone, and (2-hydroxypropyl)-β-cyclodextrin were obtained from Sigma Aldrich (St. Louis, MO, USA). Poloxamer 188 (Kolliphor®, P188), poloxamer 407 (Kolliphor®, P407), povidone K30 (Kollidon®30, PVP K30), and copovidone K28 (Kollidon® VA 64, Co-PVP) were provided by BASF-Korea (Seoul, Korea). Hydroxypropyl methyl cellulose E5 (Methocel®, HPMC E5) was obtained from Colorcon Asia Pacific PTE LTD. (Korea Branch, Suwon, Korea). Polyethylene glycol 6000 (PEG 6000) was obtained from the Daejung Chemical & Metal Co., Ltd. (Shiheung, Korea). Low-substituted hydroxypropyl cellulose (L-HPC) was purchased from Shin-Etsu (Tokyo, Japan). All the other chemicals were of analytical grade, and all the solvents were of high-performance liquid chromatography (HPLC) grade.

Flavonoids Modulate rhsTRAIL Activity

Soluble recombinant human TRAIL (rhsTRAIL) was purchased from PeproTech Inc. (Rocky Hill, NJ, USA). DMSO (dimethylsulfoxide), DMF (dimethylformamide) and Triton X-100 were obtained from Sigma Chemical Company (St. Louis, MO, USA). 5-Hydroxyflavone, 6-methoxyflavone, 6-Hydroxyflavone, 7-methoxyflavone and 7-Hydroxyflavone were purchased from Sigma Chemical Company. 6-Hydroxyflavone and 7-Hydroxyflavone were obtained by biotransformation of 6-methoxyflavone, 7-methoxyflavone, 6-Hydroxyflavone and 7-Hydroxyflavone. The flavones were dissolved in DMSO (≤50 mM) or in DMF (50 mM) to a final concentration of 0.1% (v/v) in culture media.
